NKG2D a KIR receptory a jejich role u alogenní transplantace krvetvorných buněk u pacientů s akutní myeloidní leukémií ABSTRAKT NK buňky (z anglického Natural Killer = přirození zabíječi) hrají důležitou roli v boji proti virovým infekcím či nádorovým onemocněním. Tyto imunitní buňky jsou schopné rozeznávat i buňky bez přítomných HLA (human leukocyte antigen) proteinů a na rozdíl od T lymfocytů tak dokáží eliminovat buňky, které se snaží uniknout imunitnímu dohledu internalizací těchto molekul. Toho jsou schopné například leukemické kmenové buňky, které jsou považovány za buňky zodpovědné za relaps leukemie. Aktivita NK buněk je řízena pomocí inhibičních a aktivačních receptorů. Mezi nejlépe prostudované patří KIR (killer-cell immunoglobulin-like receptor) receptory, které mohou být jak inhibiční, tak aktivační a jejichž ligandy jsou primárně HLA molekuly. Nejvýznamnějším aktivačním receptorem je NKG2D (natural killer group 2-D), jehož ligandy jsou stresem indukované proteiny MICA a MICB (MHC class I-related chain- A nebo -B) a ULBP (human cytomegalovirus Unique Long 16-binding protein). V první části této práce jsme si dali za cíl popsat, jestli česká populace odpovídá frekvencí alel a polymorfismů receptorů KIR a NKG2D a ligandů MICA a MICB dalším populacím kavkazského původu. These immune cells are also able to recognize cells without HLA (human leukocyte antigen) proteins and, unlike T cells, can eliminate cells that try to escape immune surveillance by internalizing these molecules. For example, leukaemia stem cells, which are thought to be responsible for leukaemia relapse, are able to do this. The activity of NK cells is controlled by inhibitory and activating receptors. Among the best studied are the KIR (killer-cell immunoglobulin-like receptor) receptors, which can be both inhibitory and activating and whose ligands are primarily HLA molecules. The most prominent activating receptor is NKG2D (natural killer group 2-D), whose ligands are the stress-induced proteins MICA and MICB (MHC class I- related chain-A or -B) and ULBP (human cytomegalovirus Unique Long 16-binding protein).
