Název diplomové práce: Studium exprese a toxicity derivátů katecholu v buněčné linii MCF-7 Estrogeny plní v lidském organismu mnoho důležitých funkcí. Působí vazbou na estrogenní receptory (ER) a tím regulují reprodukci, menstruační cyklus, kostní denzitu, metabolismus cholesterolu nebo mozkové funkce. Významnou úlohu mají také v rozvoji a vzniku karcinomu prsu, kde je množství exprimovaných ER využíváno jako velmi důležitý biomarker u pacientů, kteří tímto onemocněním trpí. Karcinom prsu je i navzdory neustále probíhajícímu výzkumu považován za nádor s nejvyšší úmrtností. Katecholy jsou organické sloučeniny. V lidském organismu se mohou objevovat jako metabolity při degradaci benzenu a estrogenů anebo dalších endogenních sloučenin jako jsou neurotransmitery a jejich prekurzory. Je známo, že katecholy se v organismu účastní redoxních dějů, působí antioxidačně i toxicky, narušují funkci proteinů a způsobují zlomy v DNA. Předmětem výzkumu je jejich pozitivní vliv na terapii karcinomu prsu, nicméně ten není ještě zcela prozkoumán. Předmětem této diplomové práce je studium cytotoxické aktivity u třinácti vybraných derivátů katecholu v prsní... Title of the Diploma thesis: Study of the expression and toxicity of catechol derivatives in MCF-7 cell line Estrogens perform many important functions in the human body. They act by binding to estrogen receptors (ER) and thus regulate reproduction, the menstrual cycle, bone density, cholesterol metabolism or brain function. They also play an important role in the development and onset of breast cancer, where the amount of ER expressed is used as a very important biomarker in patients suffering from this disease. Despite ongoing research, breast cancer is considered to have the highest mortality rate. Catechols are organic compounds. In the human body, they can occur as metabolites in the degradation of benzene and estrogens or other endogenous compounds such as neurotransmitters and their precursors. Catechols are known to be involved in redox processes in the body, to exert antioxidant and toxic effects, to interfere with protein function and to cause DNA strand breaks.
