Helmintózy představují významné zdravotní riziko, proto je nezbytné k diagnostice těchto onemocnění využívat efektivní a rapidní diagnostické metody, mezi které patří i molekulární metody, jež jsou shrnuty v této bakalářské práci. Úvodní kapitola rozebírá biologii významných lidských helmintóz. Následující kapitoly popisují metody izolace, vizualizace DNA a molekulární metody, které jsou využívány v testech pro diagnostiku helmintóz. V rámci této práce jsou kapitoly zabývající se molekulárními metodami rozděleny do dvou částí. První část je věnována PCR a jeho modifikacím, druhá se zaměřuje na metody izotermální amplifikace DNA, které představují praktičtější a efektivnější alternativu k PCR. U všech metod jsou diskutovány jejich výhody, nevýhody a příklady helmintóz, u kterých byly aplikovány. Závěrečná kapitola se věnuje vlivu pandemie COVID-19 na vývoj molekulárních diagnostických metod, během které byly upraveny stávající protokoly či aplikovány nové molekulární metody za účelem zvýšení efektivity. Tato bakalářská práce přináší přehled dodnes využitých molekulárních metod na diagnostiku helmintóz. Zároveň diskutuje vliv pandemie COVID-19 na rozvoj molekulární diagnostiky a tím nastiňuje možný budoucí vývoj molekulárních diagnostických metod v oblasti helmintologie. Helminthiases represent a significant health risk, therefore the use of effective and rapid diagnostic methods is essential, including molecular methods, which are summarized in presented thesis. The biology of important human helminth pathogens is summarized in the introductory part. The DNA isolation, visualisation and molecular methods are described in the following parts. The section about molecular methods is divided into two parts. The first part is focused on PCR and its modifications, the second part describes methods of isothermal DNA amplification, which represent more practical and effective alternative to PCR. The advantages, disadvantages of all methods and examples of helminthiases to which they have been applied are also discussed. The final part is about the impact of the COVID-19 pandemic on the development of molecular diagnostic methods, during which were previously established protocols modified or new molecular methods were applied to increase the efficiency. The presented thesis brings an overview of molecular methods that were used to this day for diagnosis of helminthiases.
