Tableta Ataralgin je volně dostupné léčivo napomáhající proti bolestem a zvýšené tělesné teplotě. Tableta obsahuje tři účinné látky - paracetamol, kofein a guaifenesin. Cílem této práce bylo vyvinout rychlou, účinnou, selektivní a jednoduchou UHPLC-PDA metodu vhodnou ke stanovení účinných látek v tabletě Ataralginu, která by následně sloužila ke kontrole kvality při výrobě tohoto léčiva ve farmaceutickém průmyslu. Separace probíhala na koloně Acquity UPLC BEH C18 (50 × 2,1 mm, 1,7 µm), teplota kolony byla nastavena na 40 řC, teplota na dávkovači byla 10 řC, mobilní fáze byla složena z methanolu (B) a destilované vody s přídavkem 0,1% kyseliny mravenčí (A), objem nástřiku byl 1 µl a průtok byl nastaven na 0,4 ml·min-1 . Pro měření byla nastavena podmínka gradientové eluce (čas: 0-0,5-1,5-2,5-3-5,5 min, A: 85-85-30-30-85-85 % v/v). Analýza probíhala při 275 nm a celková doba analýzy trvala 5,5 minut. Správnost (relativní chyba) byla v rozmezí 0,05 - 2,73 % a přesnost (relativní směrodatná odchylka) byla v rozmezí 2,38 - 5,79 %. Hodnoty LOD byly následující: 9,25·10-4 mg·ml- 1 pro paracetamol, 2,15·10-4 mg·ml-1 pro kofein a 7,93·10- 4 mg·ml-1 pro guaifenesin a hodnoty pro LOQ byly 3,08·10-3 mg·ml-1 pro paracetamol, 7,17·10-4 mg·ml-1 pro kofein a 2,64·10- 3 mg·ml-1 pro guaifenesin. The tablet contains three active substances - paracetamol, caffeine and guaifenesin. The aim of this thesis was to develop a rapid, effective, selective and simple UHPLC-PDA method suitable for the determination of active substances in the Ataralgin tablet which would subsequently serve for quality control during the production of this drug in the pharmaceutical industry. Separation was performed on the Acquity UPLC BEH C18 column (50 × 2.1 mm, 1.7 µm), the column temperature was set at 40 řC, autosampler temperature was 10 řC, the mobile phase was composed of methanol (B) and distilled water with the addition of 0.1% formic acid (A), sample injection volume was 1 µl and the flow of mobile phase was 0.4 ml·min-1 , gradient elution (time: 0-0.5-1.5-2.5-3-5.5 min, A: 85-85-30-30- 85-85% v/v). UV detection at 275 nm and the total analysis time was 5.5 minutes. The trueness (relative error) was in the range of 0.05 - 2.73 % and the accuracy (relative standard deviation) was in the range of 2.38 - 5.79 %.
