Objavy posledných rokov ukazujú, že porucha mitofágie sa podieľa na patofyziológii Alzheimerovej choroby (ACH) a ostatných neurodegeneratívnych ochorení. Štúdie na mozgových biopsiách pacientov s ACH, bunkových aj zvieracích modeloch ACH ukazujú, že na veku závislý pokles kapacity mitofágie významne prispieva k patológií ACH, a že hladiny proteínov súvisiacich s mitofágiou sú zmenené. Či sa však tieto zmeny odrážajú v biofluidoch jedincov s ACH, a či by proteíny účastniace sa mitofágie mohli byť potencionálnymi biomarkermi ACH, nie je známe. Cieľom diplomovej práce bolo porovnať hladinu markerov mitofágie v krvnom sére a mozgovomiechovom moku (CSF) pacientov v rôznych štádiách ACH s kognitívne zdravými kontrolami (CU) a určiť ich vzťah k miere postihnutia kognície a štandardným biomarkerom ACH (amyloid beta 42 (Ab42), celkového tau (T-tau) a tau fosforylovaného na threoníne 181 (P-tau181)). Preukázali sme, že u jedincov s ACH je narušená mitofágia, ktorá sa prejavuje zvýšenými hladinami PINK1 a BNIP3L (aktivátory mitofágie) a zníženými hladinami TFEB (hlavný regulátor lyzozomálnej biogenézy) v porovnní s CU. Tieto zmeny boli navyše spojené s pokročilejšou patológiou ACH, ktorá sa prejavila zvýšenou pozitivitou biomarkerov ACH a postihnutím kognície. Tieto výsledky naznačujú, že pri ACH dochádza k... The findings of recent years have shown that impaired mitophagy is involved in the pathophysiology of Alzheimer's disease (AD) and other neurodegenerative diseases. Studies on brain biopsies of AD patients, cellular and animal models of AD show that age-dependent decline in mitophagy is a significant contributor to AD pathology, and that the levels of mitophagy proteins are altered. However, whether these changes are reflected in the biofluids of individuals with AD, and whether mitophagy proteins could be potential biomarkers of AD, is unknown.The aim of the diploma thesis was to compare the level of mitophagy markers in blood serum and cerebrospinal fluid (CSF) of patients in various stages of AD with cognitively healthy controls (CU) and determine its relationship to the degree of cognitive impairment and standard Alzheimer's biomarkers (amyloid beta (Ab42), total tau (T-tau) and tau phosphorylated at threonine 181 (P-tau181)). We have shown that mitophagy is impaired in individuals with AD, manifested by increased levels of PINK1 and BNIP3L (activators of mitophagy) and decreased levels of TFEB (master regulator of lysosomal biogenesis) compared to CU.
