PharmDr. Petr Zimčík, Ph.D. Dizertačná práca: Výzkum látek ovlivňujících teplotu tání oligonukleotidových sond Real-time PCR je široko využívanou metódou v rôznych oblastiach výskumu ako sú napr. biomedicína, mikrobiológia, veterinárna medicína, atď. Kvantifikácia génovej expresie, alelická diskriminácia či detekcia sekvenčných variantov sú novými a zaujímavými možnosťami pre PCR. Diskriminácia bodových zmien pri použití dlhších sond nie je optimálna. Rozdiel teploty topenia medzi plne komplementárnym duplexom a duplexom obsahujúcim bodovú zmenu je v prípade použitia dlhších sond zanedbateľný. Na druhej strane, krátke oligonukleotidové (ON) sondy sú vhodné na dobrú diskrimináciu bodových zmien. Krátke ON sondy majú nízke teploty topenia. Tento problém je možné vyriešiť pripojením látok viažucich sa do malého žliabku DNA (minor groove binder, MGB), či iných látok, ktoré majú silnú interakciu s DNA ako napríklad interkalačné farbivá či polyamíny. PharmDr. Petr Zimčík, Ph.D. Doctoral Thesis: Investigation of the compounds influencing the melting temperature of oligonucleotide probes Real-time PCR is widely used method in various research fields like biomedicine, microbiology, veterinary medicine, etc. Quantification of gene expression, allelic discrimination and alteration detection are new and interesting application possibilities. Mismatch discrimination is not optimal when longer probes are used. Melting temperature difference between fully complementary duplex and mismatched duplex is negligible in the case of longer probes. The short oligodeoxynucleotide (ODN) probes, on the other hand, are suitable for good discrimination of single nucleotide variants. However, these probes suffer from low melting temperatures. Adding minor groove binders (MGB), or other substances, which can strongly interact with DNA such as intercalating dyes or polyamines, can increase the melting temperature of short probes duplexes.
