[cze][eng] Cílem této bakalářské práce bylo zjištění obsahu biologicky účinných biogenních aminů a polyaminů ve skopovém a jehněčím mase. Česká republika není příliš velkým konzumentem skopového a jehněčího masa. Nejvíce se ho konzumuje v období Velikonoc, zatímco v anglosaských zemích je vyhledávaným druhem. Spotřeba skopového masa ve světě je zhruba 2 kg/os./ rok, zatímco v ČR pouze 0,4 kg/os./ rok. Maso jatečných zvířat je bohaté na biogenní aminy a polyaminy, především na spermin (SPM) a spermidin (SPD). Jejich obsah již byl stanoven u masa a drobů skotu, prasat a kuřat. Údaje o obsahu polyaminů SPM a SPD ve skopovém i jehněčím mase dosud v literatuře chybějí, údaje o obsahu biogenních aminů jsou velmi kusé. Za použití metody kapalinové chromatografie v uspořádání UPLC byly stanovovány aminy: putrescin (PUT), kadaverin (CAD), tyramin (TYR), fenylethylamin (PEA), histamin (HIM), tryptamin (TRM), SPD a SPM, které mají výrazné biologické účinky. Analyzováno bylo maso skopové (19 vzorků z hřbetu, 17 vzorků z kýty)a jehněčí (20 vzorků z hřbetu, 20 vzorků z kýty). Vzorky pocházely od drobných chovatelů, zvířata byla hybridy několika plemen. Všechny byly analyzovány 24 hod po porážce. Nejvyšší průměrný obsah byl zjištěn ve skopové kýtě, a to SPM 32,53 mg/kg, SPD 5,91 mg/kg a PUT 2,45 mg/kg. Druhý nejvyšší průměrný obsah měla jehněčí kýta - SPM 17,25 mg/kg, SPD 4,22 mg/kg a PUT 2,04 mg/kg. Následoval skopový hřbet s obsahem SPM 17,22 mg/kg, SPD 3,99 mg/kg, PUT 2,38mg/kg a jako u jediného ze čtyř typů masa byl ve stopovém množství zjištěn i TYR. Nejnižší obsah aminů byl zjištěn v jehněčím hřbetu, kde SPM dosáhl průměrné hodnoty 16,54 mg/kg, SPD 5,08 mg/kg a PUT 2,11 mg/kg. Histamin se vyskytoval jen ve stopových množstvích, aminy TRM, PEA a CAD nebyly u žádných vzorků zjištěny. Zjištěné hodnoty jsou srovnatelné s publikovanými údaji pro maso hovězí a vepřové. The aim of the bachelor thesis was to determine content of biologically active biogenic amines and polyamines in mutton and lamb meat. There are not too many consumers of mutton in the Czech Republic. This kind of meat is consumed mostly during Easter, while in Anglo-Saxon countries it has been a sought-after food resource. World{\crq}s mutton yearly consumption has been about 2 kg per capita. In the Czech Republic it is only 0.4 kg. Meat of slaughter animals is rich in biogenic amines and polyamines, especially in spermine (SPM) and spermidine (SPD). Their contents in beef, pork and chicken meat have been determined previously. Literature data on mutton and lamb has been lacking for polyamines and information on biogenic amines has been very scarce. Liquid chromatography method in UPLC arrangement was used for the determination of putrescine (PUT), cadaverine (CAD), tyramine (TYR), phenylethylamine (PEA), histamine (HIM), tryptamine (TRM), SPD and SPM, the amines with considerable biological effects. Mutton and lamb loin (19 and 20, respectively) and leg (17 and 20, respectively) were analysed 24 hours after slaughter The samples originated from various minor breeders and they were hybrids of several breeds. The highest mean content was determined in mutton leg (32.53, 5.91 and 2.45 mg kg-1 of SPM, SPD and PUT, respectively), followed by lamb leg (17.25, 4.22 and 2.04 mg kg-1 of SPM, SPD and PUT, respectively). The respective mean contents in mutton loin were 17.22, 3.99 and 2.38 mg kg-1 and those in lamb loin 16.54, 5.08 and 2.11 mg per kg. Lamb loin was the only meat with detected TYR. Histamine was present only at trace levels, amines TRM, PEA and CAD were not detected in any samples. The determined contents are comparable with published data for beef and pork.
