Práca sa zameriava na zistenie vplyvu vybraných typov potravín na zmeny kompozície črevného mikrobiómu. Zloženie stravy hrá dôležitú úlohu pri kontrole črevných mikrobiálnych populácií, vrátane prevencie a liečbe určitých ochorení. S cieľom vyhodnotiť vplyv dennej konzumácie ovocno-zeleninového smoothie na ľudský črevný mikrobióm boli porovnané zmeny v črevnom mikrobióme na začiatku a po 14 dňoch intervencie. Experimentálnej časti sa zúčastnilo 10 mužov a 10 žien vo veku 35-45 rokov, ktorí boli rovnomerne rozdelení do 2 skupín. Prvá skupina účastníkov konzumovala kombináciu zloženú z pomaranču, sušenej kurkumy a zázvoru. Druhá skupina konzumovala kombináciu zloženú z jablka, mrkvy a citrónovej šťavy. Na zistenie rozdielov v črevnom mikrobióme bola využitá izolácia DNA zo vzoriek stolice a následne kvantitatívna PCR metóda. Po 14-dňovej konzumácií ovocno-zeleninového smoothie boli pozorované určité zmeny kmeňov Firmicutes, Bacteroidetes a rodov Lactobacillus, Bifidobacterium. Po štatistickom vyhodnotení dosiahnutých výsledkov neboli rozdiely preukázateľné na štatisticky významnej hladine (p-hodnota > 0,05). The master's thesis focuses on determining the influence of selected types of food on changes in the intestinal microbiome composition. Dietary composition plays an important role in controlling intestinal microbial populations, including the prevention and treatment of certain diseases. In order to evaluate the effect of daily consumption of fruit-vegetable smoothie on the human intestinal microbiome, changes in the intestinal microbiome at the beginning and after 14 days of intervention were compared. The experimental part was attended by 10 men and 10 women aged 35-45 years, who were evenly divided into 2 groups. The first group of participants consumed a combination of orange, dried turmeric and ginger. The second group consumed a combination of apple, carrot and lemon juice. DNA isolation from stool samples followed by a quantitative PCR method was used to detect differences in the intestinal microbiome. After 14 days of consumption of the fruit-vegetable smoothie, specific changes in the phylum Firmicutes, Bacteroidetes and the genus Lactobacillus, Bifidobacterium were observed. After statistical evaluation of the obtained results, the differences were not detectable at a statistically significant level (p-value > 0.05).
