Tato bakalářská práce se v úvodu věnuje stručnému přehledu základních zobrazovacích metod používaných v klasické a kvantitativní fázové mikroskopii. Hlavním cílem práce je navrhnout, zkonstruovat a odzkoušet tomografický osvětlovač kompatibilní s laboratorní verzí mikroskopu Q-PHASE. Konstrukční řešení je rozloženo do několika částí, kdy v prvním kroku jsou stanoveny optické požadavky soustavy pomocí zákonů geometrické optiky. Následně jsou vybrány vhodné optické komponenty a v softwaru Zemax provedeny simulace k ověření, že jsou všechny stanovené požadavky splněny. Při konstrukci je dbáno na kompaktnost a kompatibilitu sestavy. Proto je k osvětlení amplitudového modulátoru světla použit hranol s totálním vnitřním odrazem. Dále jsou navrhnuta konstrukční řešení uložení zdroje světla, hranolu a amplitudového modulátoru světla. Vypracovaná výkresová dokumentace je přílohou této práce. Celá sestava je složena, seřízena a pomocí 16 testovacích tomografických projekcí ověřena její správná funkce. This bachelor thesis starts with a brief overview of the basic imaging methods used in classical and quantitative phase microscopy. The main aim of the thesis is to design, construct and test a tomographic illuminator compatible with the laboratory version of the Q-PHASE microscope. The design is decomposed into several parts, where in the first step the optical requirements of the system are determined using the laws of geometrical optics. Suitable optical components are then selected and simulations are performed in the Zemax software to verify that all specified requirements are met. The compactness and compatibility of the assembly is taken into account in the design. Therefore, a prism with total internal reflection is used to illuminate the amplitude light modulator. Furthermore, the design solutions for mounting the light source, the prism and the amplitude modulator are proposed. The developed drawings are attached to this thesis. The whole assembly is assembled, adjusted and its correct function is verified by means of 16 test tomographic projections.
