Nádorová onemocnění patří mezi nejzávažnější a zároveň nejčastější choroby současné populace. Z toho důvodu jsou prováděny výzkumy, jejichž cílem je objevit nové způsoby léčby rakoviny. Jedním z často používaných léčebných postupů je chemoterapie. Nicméně kvůli nežádoucím účinkům na zdraví onkologických pacientů současně známých chemoterapeutik jsou stále vyvíjeny nové látky s protinádorovým účinkem. V této bakalářské práci byly srovnány postupy mineralizace nádorových buněk, do nichž byla aplikována chemoterapeutika. Jedním z rozkladných přístupů byla mineralizace na mokré cestě s použitím kyseliny dusičné a peroxidu vodíku v různých poměrech. Tento způsob mineralizace byl srovnán s mineralizací pomocí mikrovlnného zařízení, kde byla mineralizace nejúčinnější. Mineralizace nádorových buněk totiž byla prvním krokem ve zjištění účinnosti chemoterapeutika v určité buněčné linii. Pro analýzu nádorových buněk byla použita metoda ICP-MS, před kterou bylo potřeba vzorky buněk převést do roztoku, a to bylo provedeno právě mineralizací. Bohužel kvůli variabilitě použitých biologických vzorků nebylo možné jednoznačně posoudit, zda peroxid vodíku obsažený v rozkladné směsi přispěl ke zvýšení účinnosti mineralizace. Cancer is one of the most serious and most common diseases in today's population. For this reason, research is being carried out to discover new ways of treating cancer. One of the frequently used treatments is chemotherapy. However, due to the side effects on the health of cancer patients of the currently known chemotherapeutic agents, new substances with anticancer effect are still being developed. In this bachelor thesis, the mineralization processes of cancer cells injected with chemotherapeutic agents were compared. One of the digestion approaches was wet digestion using nitric acid and hydrogen peroxide in different ratios. This method of digestion was compared with digestion using a microwave device, where mineralization was most efficient. Indeed, mineralization of tumor cells was the first step in determining the efficacy of a chemotherapeutic drug in a particular cell line. The ICP-MS method was used to analyse the tumour cells, prior to which the cell samples had to be transferred into solution and this was done by digestion. Unfortunately, due to the variability of the biological samples used, it was not possible to clearly assess whether the hydrogen peroxide contained in the digestion mixture contributed to the efficiency of mineralisation.
