Teoretická část práce pojednává o široké škále mikroorganismů a látek odvozených od probiotik. V práci jsou na základě nejnovějších publikací definovány probiotika, prebiotika, synbiotika, paraprobiotika, postbiotika, pharmabiotika, psychobiotika a onkobiotika. Dále je specifikován účel a použití zmíněných – biotik k současnému poznání jejich funkce. V experimentální části byla ze vzorku probiotických tobolek izolována DNA komerčně dostupným bakteriálním kitem a magnetickými částicemi. Byla poté spektrofotometricky stanovena její čistota a koncentrace. Za použití izolované DNA jako matrice pro konvenční PCR a detekce na agarózovém gelu bylo analyzováno složení probiotického produktu deklarovaného výrobcem. V závěru byla optimalizována metoda PMA-PCR. The theoretical part of the thesis deals with a wide range of microorganisms and substances derived from probiotics. Based on recent publications, probiotics, prebiotics, synbiotics, paraprobiotics, postbiotics, pharmabiotics, psychobiotics and oncobiotics are defined. Furthermore, the purpose and use of the mentioned -biotics is specified for the current understanding of their function. In the experimental part, DNA was isolated from a sample of probiotic capsules using a commercially available bacterial kit and magnetic particles. Its purity and concentration were then determined spectrophotometrically. Using the isolated DNA as a matrix for conventional PCR and detection on agarose gel, the composition of the probiotic product as declared by the manufacturer was demonstrated. Finally, the PMA-PCR method was optimized.
