Nedílnou součástí života buněk je buněčná signalizace, která je zajištěna mimo jiné hemovými proteiny, konkrétně jejich skupinou s názvem hemové senzorové proteiny. Hemové senzorové proteiny se dělí do dvou skupin, hemové senzorové proteiny detekující hem, které se vyskytují spíše v eukaryotních buňkách a hemové senzorové proteiny detekující plynné molekuly, které jsou typické spíše pro prokaryotní buňky. Plynnými molekulami mohou být v tomto případě CO, NO a O2. Skupina hemových senzorových proteinů detekujících kyslík má kruciální význam pro bakterie, které se musí přizpůsobovat proměnlivé koncentraci kyslíku v okolním prostředí. Jednou z těchto bakterií je půdní bakterie Anaeromyxobacter sp., kmen Fw 109-5, která obsahuje hemový senzorový protein s globinovou strukturou senzorové domény a histidinkinasovou aktivitou domény funkční (AfGcHK), umožňující bakteriím kromě jiného například tvorbu biofilmu.. . V rámci teoretické části této práce byly shrnuty dosavadní poznatky o hemových senzorových proteinech se zaměřením na protein AfGcHK. V praktické části této bakalářské práce byly transformovány bakteriální buňky E. coli BL-21 (DE3) plazmidem pET21(+)/AfGcHK obsahujícím gen kódující AfGcHK, ze kterých byly následně izolovány dvě formy AfGcHK, přirozená forma a forma obsahující analog hemu,... An integral part of the life of cells is cell signaling, which is ensured, among other things, by heme proteins, specifically by their members called heme sensor proteins. Heme sensor proteins are divided into two groups, heme sensor proteins detecting heme, which are found more in eukaryotic cells, and heme sensor proteins detecting gaseous molecules, which are more typical of prokaryotic cells. The gas molecules in this case can be CO, NO and O2. A family of oxygen-detecting heme sensor proteins is crucial for bacteria that must adapt to changing oxygen concentrations in the environment. One of these bacteria is the soil bacterium Anaeromyxobacter sp., strain Fw 109-5, which contains a heme sensor protein with the globin structure of the sensor domain and the histidine kinase activity of the functional domain (AfGcHK), enabling the bacteria, among other things, to form a biofilm. As part of the theoretical part of this thesis, the current knowledge about heme sensor proteins was summarized, with a focus on the AfGcHK protein.
