[eng][cze] In this thesis, we describe the synthesis and self-assembly of new photo-responsive triblock terpolymers by RAFT polymerization. These triblock terpolymers were characterized by GPC, NMR and IR spectroscopy. The resulting nanoparticles were characterized by DLS, SAXS and Cryo-EM. To avoid problems with RAFT polymerization of the photo-responsive monomer o- nitrobenzyl acrylate, we used naphthacyl methacrylate, a new RAFT-polymerizable photo- responsive monomer. In addition, we used phenylboronic acid moieties to conjugate model drugs with our triblock terpolymers via dynamic covalent bonding and studied stimuli-triggered release of a model drug-like compound, Alizarin Red S. Our results showed that naphthacyl methacrylate is indeed polymerizable by RAFT and that such polymers retain photo-responsive properties. We were also able to bind Alizarin Red S to the polymer via boronate ester linkage and trigger its release by dopamine or fructose addition, both of which are biologically relevant compounds. Therefore, our photo- and diol-responsive nanoparticles open up opportunities for developing new drug-delivery systems.V diplomové práci je popsána syntéza nových fotoresponzivních triblokových terpolymerů pomocí polymerizace typu RAFT a dále samouspořádání výsledných terpolymerů ve vodných roztocích. Triblokové terpolymery byly charakterizovány pomocí gelové permeační chromatografie, NMR a IR spektroskopie. Připravené nanočástice byly charakterizovány pomocí rozptylových metod DLS a SAXS a mikroskopické techniky Cryo-EM. Jelikož se běžně používaný fotocitlivý monomer o-nitrobenzylakrylát nehodí k polymerizacím typu RAFT, byl místo něho testován dosud málo využívaný naftacyl methakrylátový monomer, který je také fotocitlivý. Naše výsledky jednoznačně prokázaly, že naftacyl methakrylát je polymerizovatelný metodou RAFT a že takto připravené polymery si zachovávají fotoresponzivní vlastnosti. Kromě toho byl při syntéze triblokových terpolymerů využit blok obsahující fenylboronové kyseliny pro konjugaci modelových léčiv prostřednictvím dynamické kovalentní vazby na polymerní řetězec. V této souvislosti bylo studováno řízené uvolňování modelové sloučeniny Alizarin Red S z polymerních nanočástic vyvolané přítomností dopaminu a fruktózy. Tyto nanočástice, detailně studované v předložené diplomové práci, reagující na světlo a dioly, otevírají nové možnosti pro vývoj sytémů vhodných pro cílený transport léčiv.
