The ability to sense painful stimuli is essential to protect the body. Up to date, the underlying molecular mechanisms are still not completely understood; therefore, the treatment of chronic inflammatory pain remains challenging. The TRPV1 channel is one of the known nociceptors mediating the sensation of burning stimuli. Its agonist is capsaicin, the pungent compound of chilli peppers. This channel has been extensively studied in neurones; however, its function in immune cells is not well understood. Especially in macrophages, data regarding the role of TRPV1 in macrophage polarisation are often contradictory. Thus, further research in this area is desired to clarify the function of TRPV1 in immune cells. This diploma thesis aims to investigate the role of TRPV1 in macrophage polarisation during the inflammatory response. In this work, macrophages were stimulated with capsaicin prior to, after or concurrently with the application of LPS to determine the effect of TRPV1 activation on the inflammatory response. The involvement of MAP kinases in signalling after TRPV1 activation by capsaicin was addressed confirming that ERK 1/2 is part of the signalling cascade. Schopnost rozpoznat bolestivé podněty je zásadní pro ochranu organismu. Do dnešní doby jsou však molekulární mechanismy stojící za tímto procesem nepochopené. Z tohoto důvodu zůstává léčba chronické zánětlivé bolesti velmi obtížná. TRPV1 kanál je jedním ze známých nociceptorů, které zprostředkovávají pocit pálivé bolesti. Agonistou tohoto kanálu je kapsaicin, pálivá složka chilli papriček. TRPV1 byl důkladně studován v nervových buňkách, avšak jeho funkce v buňkách imunitního systému není stále objasněna. Obzvlášť data týkající se role TRPV1 v polarizaci makrofágů jsou často protichůdná. Pro objasnění funkce TRPV1 v imunitních buňkách je zapotřebí dalšího výzkumu. Tato diplomová práce si klade za cíl prozkoumat roli TRPV1 v polarizaci makrofágů při zánětlivé odpovědi. V této práci byly makrofágy stimulované kapsaicinem před, po anebo zároveň s aplikací LPS za účelem určení efektu aktivace TRPV1 při zánětlivé odpovědi. Bylo pozorováno zapojení MAP kináz do signalizace po aktivaci TRPV1 kapsaicinem. Potvrdilo se, že ERK 1/2 je součástí signalizační kaskády. Tato práce také poukázala, že může docházet k přesmyku mezi prozánětlivým a protizánětlivým fenotypem v důsledku aktivace TRPV1 při LPS indukované zánětlivé odpovědi, tento přesmyk závisí na kultivačních podmínkách.
