Nádorové buňky exprimující zvýšené hladiny některých ABC transportérů (hlavně P- glykoproteinu) se stávají rezistentními vůči mnoha často i strukturně a funkčně odlišným léčivům. Tato tzv. mnohočetná léková rezistence může být významnou překážkou pro úspěšnou chemoterapii maligních onemocnění. V současné době je známo značné množství nízkomolekulárních látek schopných potentně inhibovat P-glykoprotein, ale žádná z nich se zatím neuplatňuje v klinické praxi. STAT3 je transkripčním faktorem důležitým pro mnohé fyziologické procesy, nicméně jeho trvalá aktivace může vést k maligní transformaci a v nádorových buňkách se podílet na snížení citlivosti k chemoterapii. STAT3 je tudíž jedním z potenciálních cílů pro nově vyvíjená protinádorová léčiva. Inhibice STAT3 signální dráhy vede ke snížení proliferace nádorových buněk a jejich zvýšené citlivosti k indukci apoptózy. Ve výzkumu nových přístupů k léčbě nádorových onemocnění je věnována velká pozornost zvýšení efektivity a snížení nežádoucích účinků konvenčních cytostatik použitím nanomateriálů a systémů pro dopravu léčiv do nádorové tkáně. Jedním z nich jsou nosiče založené na kopolymerech N-(2- hydroxypropyl)methakrylamidu (HPMA kopolymery). V rámci této diplomové práce bylo hlavním cílem vyhodnotit efektivitu derivátů několika vybraných inhibitorů HIV... Tumor cells expressing high levels of some ABC transporters (mainly P-glycoprotein) can become resistant to many structurally and functionally different drugs. Such multidrug resistance can be a significant barrier for a successful chemotherapy of malignant diseases. There is a considerable amount of small-molecular-weight compounds capable of potent inhibition of P-glycoprotein, but none of them are approved for the clinical use. STAT3 is a transcription factor important for many physiological processes, but its constitutive activation may lead to the malignant transformation and chemotherapy resistance in tumor cells. This molecule is thus potential target for anticancer drugs. The inhibition of STAT3 signaling should lead to lower cancer cell proliferation and their increased susceptibility to induction of apoptosis. Considerable attention is given to increase the effectiveness and to lower the adverse effects of conventional cytostatic agents via using nanomaterials and drug delivery systems in the research of new cancer therapy approaches. Polymeric carriers based on N-(2-hydroxypropyl)methacrylamide (HPMA) copolymers are promising candidates in this field.
