[cze][eng] Cílená protinádorová terapie je novodobým efektivním přístupem k léčbě nádorových onemocnění. Tento typ léčby zahrnuje několik skupin protinádorových léčiv. Druhou nejčastěji využívanou skupinou cílených chemoterapeutik jsou inhibitory tyrosinkinas. Jedná se o nízkomolekulární látky, které cíleně zasahují do určitých signalizačních drah nádorové buňky a tím inhibují její růst, proliferaci a angiogenezi. Zástupci této skupiny léčiv jsou např. vandetanib a sunitinib. Vandetanib se primárně využívá k léčbě medulárního karcinomu štítné žlázy a sunitinib byl schválen pro léčbu gastrointestinálního stromálního tumoru a pro léčbu pokročilého renálního karcinomu. Předkládaná diplomová práce se zabývá vlivem pH a teploty na in vitro metabolismu sunitinibu a vandetanibu pomocí enzymů v první fázi biotransformace. Při experimentech byly použity různé jaterní mikrozomy z premedikovaných a kontrolních potkanů. Pro porovnání byly využity lidské a potkaní rekombinantní cytochromy P450. Primárním účelem experimentů bylo nalezení experimentálního pH a teplotního optima jejich in vitro metabolismu. V rámci těchto experimentů byl zkoumán i vliv pH a teploty na samotná léčiva a jejich stabilitu. Ovlivnění metabolismu vandetanibu a sunitinibu pomocí pH a teploty bylo analyzováno pomocí vysokoúčinné kapalinové...Targeted anti-tumor therapy is a modern and effective approach to cancer treatment. This type of therapy includes several groups of anticancer drugs. The second most commonly used group of targeted chemotherapeutic substances are tyrosinkinase inhibitors. These are small- molecular agents that target specific signalling pathways of the tumor cell, thereby inhibiting it's growth, proliferation and angiogenesis. Representatives of this drug group include vandetanib and sunitinib. Vandetanib is primarily used for the treatment of medullary thyroid cancer and sunitinib has been approved for the treatment of gastrointestinal stromal tumor and advanced renal cell carcinoma. The diploma thesis focuses on the effect of pH and temperature on the in vitro metabolism of vandetanib and sunitinib using first-phase biotrasnformation enzymes. Differently premedicated and control rat liver microsomes were used in the experiments. Human and rat recombinant cytochromes P450 were used for comparison. The primary purpose of the experiments was to find the experimental pH and temperature optimum of the in vitro metabolism of vandetanib and sunitinib.
