Kolicin U je protein produkovaný bakterií Shigella boydii. Řadí se do skupiny pórotvorných kolicinů, které interagují s receptory ve vnější membráně bakterií blízce příbuzných produkčnímu kmeni. Následně je kolicin translokován přes vnější membránu a periplazmu. Nakonec se váže do cytoplazmatické membrány, kde tvoří transmembránové póry, které tuto membránu depolarizují, a v důsledku pak způsobují zánik buňky. Pórotvorné vlastnosti kolicinu U zatím nebyly podrobně studovány, proto jsem se ve své práci rozhodla tímto tématem zabývat. Ukázalo se, že póry kolicinu U jsou pravděpodobně tvořeny jedinou molekulou kolicinu a jsou napěťově ovládané. Kromě toho, měřením s neelektrolyty bylo možné stanovit přibližný vnitřní průměr póru (0,7-1 nm) a také byl určen jeho teoretický vnitřní profil. V neposlední řadě se tato práce zabývá uspořádáním membránových α-helixů pórotvorné domény kolicinu U v otevřeném stavu. S využitím biotinylačního značení bylo zjištěno, že po otevření kolicinového póru, dochází k translokaci části pórotvorné domény na opačnou stranu membrány. Konkrétně se ukázalo, že aminokyseliny na pozici F463 a D486 (mezi α-helixy H2-H4) jsou po otevření póru přítomné na trans straně membrány. Na závěr se tato práce také zabývá vlastnostmi peptidu H1, který odpovídá části prvního α-helixu... Colicin U is a protein produced by bacterium Shigella boydii. It belongs to the group of pore-forming colicins. These colicins interact with receptors in the outer membrane of bacteria closely related to a producing colicinogenic strain. After interaction with the receptor, colicin is translocated across the outer membrane and periplasm to the cytoplasmic membrane where it forms pores. Consequently, the pore formation leads to membrane depolarization and cell death. In this thesis I decided to study the pore-forming properties of colicin U and its membrane topology. It is shown that colicin U pores are formed by only one colicin molecule and they are voltage dependent. Using measurements with nonelectrolytes we estimated a theoretical inner profile of the pore and its inner diameter to be between 0.7 and 1 nm. Above that, a membrane topology of colicin U pore-forming domain (PFD) is studied. BLM measurements with biotinylated colicin U showed that a significant part of colicin's PFD was translocated to the opposite side of the membrane after the pore opening. The segment between substituted amino acids F463 and D486 was evidenced to be on the trans side of the membrane after the pore opening.
