Doposud nebyla nalezena léčba, která by zcela zastavila progresi Alzheimerovy nemoci. Dříve používaný takrin měl v terapii Alzheimerovy choroby dobré výsledky, nicméně dlouhodobé užívání vedlo k chronické hepatotoxicitě způsobené jeho metabolity. Tato diplomová práce se zabývá sloučeninou 7-fenoxytakrin, která je jedním z nadějných substituovaných derivátů takrinu a mohla by být jedním z kandidátů pro potenciální využití v terapii Alzheimerovy choroby. Z důvodu vzniku hepatotoxických metabolitů takrinu po biotransformaci v játrech se jeví jako nezbytné u molekuly 7-fenoxytakrinu identifikovat jeho vznikající metabolity. V rámci diplomové práce byla provedena in vitro biotransformační studie 7-fenoxytakrinu za použití lidských jaterních mikrosomů. Pro stanovení parentní látky a vznikajících sedmnácti metabolitů byla využita technika vysokoúčinné kapalinové chromatografie spojená s tandemovou hmotnostní spektrometrií. Výsledky ukázaly vznik šesti monohydroxylovaných a jedenácti dihydroxylovaných metabolitů 7-fenoxytakrinu. Hydroxylace je tedy hlavní metabolická reakce po in vitro mikrosomální biotransformaci. Kromě identifikace metabolitů byla v rámci této diplomové práce provedena i kvantifikační a mikrosomálně stabilitní studie zahrnující stanovení úbytku parentní látky v čase. Dále byl stanoven... Recently used tacrine showed good results in the treatment of Alzheimer's disease, however long-term use led to chronic hepatotoxicity due to its metabolites. This master thesis deals with the compound 7-phenoxytacrine, one of the promising tacrine derivatives, which is one of the candidates for potential use in the therapy of Alzheimer's disease. Due to the formation of hepatotoxic metabolites of tacrine after the biotransformation in human liver, it appears necessary to identify the emerging metabolites of 7-phenoxytacrine molecule. Within this master's thesis in vitro biotransformation study of 7-phenoxytacrine using human liver microsomes was performed. High performance liquid chromatography with tandem mass spectrometry was used to determine the parent substance and the seventeen 7-phenoxytacrine metabolites. The analytical method showed the formation of six monohydroxylated and eleven dehydroxylated metabolites of 7-phenoxytacrine. Thus, we concluded that hydroxylation is the major metabolic reaction after in vitro microsomal biotransformation.
