[cze][eng] Hydrogely patří mezi biomateriály užívané k rekonstrukci experimentálního poranění míchy (spinal cord injury, SCI). V modelu akutního SCI jsme im-plantovali hydrogely na bázi 2-hydroxyetylmetakrylátu (HEMA) a hydroxy-propylmetakrylamidu (HPMA). Po 1 měsíci hydrogely přemostily posttrau-matickou kavitu, dobře adherovaly k míšní tkáni a vytvořily permisivní pro-středí, kam vrůstaly cévy, axony a Schwannovy buňky. Fyzikální modifikace (např. povrchový náboj) hydrogelů mohou zlepšit přemostění akutního SCI. V hemisekci míšní jsme implantovali hydrogely na bázi HEMA s povrchovými náboji a porovnali s hydrogelem bez náboje. Hydrogely s povrchovým nábojem po 1 měsíci podpořily adhezi pojivových buněk a růst axonů do implantátu v porovnání s hydrogelem bez náboje. Vstřebatelné hydrogely mohou přemostit míšní lézi a následně se vstře-bat. V modelu akutního SCI jsme implantovali hydrogely na bázi kopolyme-ru HPMA a etoxyetylmetakrylátu (EOEMA), které se vstřebávaly směrem z periferie, jež byla po 1 měsíci nahrazena novou tkání, směrem do centra, tvořeného amorfní hmotou zbytku hydrogelu s minimem vrostlé tkáně. Přemostění míšní léze můžeme zlepšit odloženou implantací hydrogelu. V transekci míchy jsme implantovali hydrogel na bázi HEMA akutně a od-loženě (po 7 dnech). Odložená implantace redukovala objem...Hydrogels are biomaterials used in the treatment of experimental spinal cord injury (SCI). In a model of acute SCI, we implanted hydrogels based on 2-hydroxyethylmethacrylate (HEMA) and hydroxypropylmethacryla-mide (HPMA). One month after implantation the hydrogels bridged the cavity, adhered well to the spinal cord and created permissive environment, infiltrated with blood vessels, axons and Schwann cells. Physical modifications (e.g. surface charge) of hydrogels may improve bridging of acute SCI. We implanted hydrogels based on HEMA with a surface charge in spinal cord hemisection and compared with a hydrogel without charge. Hydrogels with surface charge improved connective tissue adhesion and growth of axons compared to a hydrogel without charge. Biodegradable hydrogels may bridge a lesion followed by complete re-sorption. In a model of acute SCI we implanted hydrogels based on the copolymer of HPMA and etoxyethylmethacrylate (EOMA) degraded from the periphery, which was substituted with new tissue after 1 month, to the center, comprising amorphous residuals of the hydrogel.
