1 Souhrn Název: Mechanizmy účinku moderních antiepileptik (hodnocení nežádoucích účinků) Autor: Lucie Butorová Vedoucí diplomové práce: Doc. MUDr. Josef Herink, DrSc. Katedra: Katedra biologických a lékařských věd Epilepsie je časté neurologické onemocnění. Je charakterizováno spontánně se opakujícími epileptickými záchvaty, které jsou způsobeny abnormalitami elektrické aktivity mozku. Terapie epilepsie spočívá v profylaxi epileptických záchvatů, a proto je nutné dlouhodobé užívání antiepileptik, které s sebou nese riziko výskytu nežádoucích účinků. Diplomová práce klasifikuje tyto epileptické záchvaty, popisuje patofyziologii epilepsie a principy terapie antiepileptiky. Hlavním cílem mé diplomové práce bylo pomocí dotazníku zjistit a vyhodnotit nežádoucí účinky antiepileptik. Výsledky potvrdily větší úspěšnost a méně nežádoucích účinků monoterapie ve srovnání s kombinovanou terapií a také moderních antiepileptik ve srovnání s 1. generací antiepileptik. V závěru bylo mou snahou srovnat úspěšnost a výskyt nežádoucích účinků u jednotlivých antiepileptik a dále pak srovnat tyto výsledky s publikovanými daty o jednotlivých antiepilepticích. 1 Summary Title: Mechanisms of effect of new antiepileptic drugs (An evaluation of adverse effects) Author: Lucie Butorová Supervisor: Doc. MUDr. Josef Herink, DrSc. Department: Department of biological and medicinal sciences Epilepsy is frequent neurological disease. It is characterised by spontaneously repeated epileptic seizures, which are caused by abnormalities of neuronal activity of the brain. Therapy of epilepsy consists in prophylaxis of epileptic seizures, and that is why it is necessary to use the antiepileptic drugs for a long time. This administration is accompanied by the increased risk of adverse effects. Present thesis classifies particular epileptic seizures, describes pathophysiology of epilepsy and the principles of therapy with antiepileptic drugs. The principal aim of my work was to find out and evaluate side effects of antiepileptic drugs by the help of the questionnaire. The results proved higher efficacy and fewer adverse effects of monotherapy in a comparison with combined therapy and also 3rd generation antiepileptic drugs in a comparison with the 1st generation of antiepileptic drugs.
