Jeden z hlavních účinků prozánětlivých cytokinů je indukce chemokinů a exprese adhezivních molekul, které regulují migraci imunitních buněk do centra poškození. Chemoatraktivní gradient zajišťuje kromě toho také fyziologický přísun buněk do tkání a lymfatických orgánů za normálních okolností. Chemokiny jsou chemotaktické cytokiny, které tvoří velmi rozsáhlou a rozmanitou skupinu vylučovaných proteinů, které mají mnoho funkcí jednak v procesech udržujících homeostázu, ale i ve stavech zánětlivých. Produkce některých chemokinů má také zásadní vliv na odhojení štěpu. Další pochopení mechanismů, kterými se účastní chemokiny akutní rejekce, by mohlo přispívat ke zlepšení léčebných kroků v transplantologii. V této diplomové práci byly sledovány sérové hladiny chemokinů u pacientů s transplantovanou ledvinou, tato měření však neukázala významnou dynamiku. Dále byl sledován vliv efektu prozánětlivých cytokinů na uvolnění chemokinů z buněk renálního epitelu a monocytů. Byly prováděny experimenty, kde se sledovaly hladiny jednotlivých chemokinů jako ENA-78, IL-8, MCP-1, MIP-1 β, RANTES, GRO alpha, u buněčných kultur THP-1 (monocyto/makrofágová buněčná linie), RPTEC (renální epiteliální buňky proximálních tubulů) a RA (nádorová linie ledvinových buněk). Further understanding of the mechanisms involved in the acute rejection chemokine could contribute to improving treatment steps in transplantology. In this diploma thesis, serum chemokine levels were monitored in renal transplant patients, but these measurements did not show significant dynamics. Furthermore, the effect of pro-inflammatory cytokines on the release of chemokines from renal epithelial cells and monocytes was studied.
