PharmDr. Jitka Mužíková, Ph.D. Posluchač: Thao Tranová Název diplomové práce: Vliv typu plniva a rozvolňovadla na vlastnosti tablet s vysokodávkovým léčivem Práce se zabývá studiem vlivu plniv a extragranulárních rozvolňovadel na vlastnosti tablet s vysokodávkovým léčivem. Ve vysokosmykovém granulátoru byly vyrobeny dva typy granulátů lišící se mikrokrystalickou celulosou a laktosou. Následně z nich bylo připraveno jedenáct skupin tablet s rozdílným typem a množstvím rozvolňovadla. Tablety obsahovaly 2 %, 3,7 % nebo 5,4 % sodné soli kroskarmelosy nebo 2 % a 5,4 % krospovidonu. Testovanými parametry tablet byly oděr, pevnost, doba rozpadu a pyknometrická hustota. Zrychlené stabilitní studie trvající 6 měsíců byly provedeny při teplotě 40 řC a relativní vlhkosti 75 %. Tablety byly průběžně hodnoceny po 1,5, 3 a 6 měsících. Všechny tablety měly v čase T0 odpovídající oděr, pevnost, dobu rozpadu a pyknometrickou hustotu. Tablety obsahující mikrokrystalickou celulosu a sodnou sůl kroskarmelosy se rozpadly rychleji než tablety s laktosou nebo krospovidonem. PharmDr. Jitka Mužíková, Ph.D. Student: Thao Tranová Title of Thesis: Influence of fillers' and disintegrants' type on properties of tablets with high dose drug The work engages in the study of fillers' and extragranular superdisintegrants' influence on properties of tablets with high dose drug. Two types of granulate with lactose or microcrystalline cellulose were produced in high shear granulator. Eleven tablets groups with different type and amount of superdisintegrants were prepared from the granulates. Tablets contained 2 %, 3.7 %, 5.4 % of sodium croscarmellose or 2 % and 5.4 % of crospovidone. The tested parameters of tablets were friability, hardness, disintegration time and density. Accelerated stability studies lasting 6 months at temperature of 40 řC and relative humidity 75 % were done. The tablets were evaluated after 1.5, 3 and 6 months. All tablets had adequate friability, hardness, disintegration and density at T0. Increasing concentration of superdisintegrant decreased disintegration time.
