Viry Zika a Dengue kódují vlastní enzymy, které jsou aktivní v různých stádiích replikačního cyklu. NS3, NS5 protein a jejich kofaktory hrají nepostradatelnou roli ve fungování flavivirů. I když byly struktury těchto proteinů již vyřešeny, jejich vlastnosti jsou prozkoumány jen částečně a mnoho aspektů ohledně jejich funkce zůstává nevyřešeno. Hlavními tématy této práce je úloha NS3 a NS5 proteinu v životním cyklu virů Zika a Dengue, sestřih polyproteinového prekurzoru, samotná replikace a interakce těchto enzymů s dalšími strukturními a nestrukturními proteiny. NS3 a NS5 protein mají mnoho enzymatických aktivit strukturně i funkčně oddělených, což je činí zajímavými z hlediska výzkumu autoaktivace, interakce funkčních domén a proteinů mezi sebou. Jelikož jsou infekce viry Zika a Dengue stále velmi vážným problémem, je nezbytné co nejlépe pochopit molekulární mechanismy jejich replikace. Klíčová slova: proteasa, polymerasa, Zika, Dengue, sestřih polyproteinů, protivirová terapie Zika and Dengue viruses codes their own enzymes which helps them in different stages of the replication cycle. NS3 a NS5 proteins and their cofactors play an essential role in flaviviral life cycle. Although their structure was already solved, many aspects of their function remain unclear. The main subject of this bachelor thesis is the role of these proteins in flaviviral life cycle, polyprotein cleavage, replication and protein-protein interaction. These enzymes keep many particular enzymatic activities such as protease, helicase, methyltrasferase and polymerase. They are both structurally and functionally separated, which is interesting regarding autoactivation and protein-protein interaction. Since Zika and Dengue infections remain a serious health care issue, it is necessary to understand the molecular mechanisms behind their replication.
