V ÚEB AVČR byl připraveny nové konjugáty PTX s peptidy, které jsou agonisty gonadotropního hormonu (GnRH). V předešlé studii bylo zjištěno, že některé z těchto konjugátů vykazují vyšší či srovnatelný antiproliferační účinek v porovnání se samotným PTX na prsní nádorové buňky, které mají receptory pro GnRH. Cílem této diplomové práce bylo porovnání účinku PTX a jeho nových derivátů v buňkách, které receptory pro GnRH nemají. Pro experiment byly zvoleny dva modelové systémy - buněčná linie HCT 8 odvozená z kolorektálního adenokarcinomu a primární kultura hepytocytů potkana. Ani jedna z těch linií neobsahuje receptory pro GnRH. Výsledky byly hodnoceny třemi různými metodami (MTT, NRU a x-Celligence). Testované konjugáty MP 264, MP 211 a MP 260 vykazovali nižší antiproliferační účinek na buněčné linii HCT 8 a deriváty MP 264 a MP 265 nižší cytotoxicitu v primární kultuře hepatocytů ve srovnání se samotným PTX. In Institute of experimental botany in Prague were preparated new conjugates with peptide agonist of receptors for gonadotropin-releasing hormon (GnRH). The previous studies showed that this conjugate have higher or same antiproliferative effect. This antiproliferative effect was compared with PTX on the cell line from breast cancer which have receptors for GnRH. The aim of this study was to compare the anti-proliferativ effect of PTX and its conjugates. For the experiment was chosen to two cell systemes - HCT 8, which is derivate from colorectal adenocarcinoma and primary culture from liver cell from sewer-rat. Both cell lines have not receptors for GnRH. The results were evaluated by three different methods (MTT, NRU and x-Celligence). The conjugates MP 264, MP 211 and MP 260 showed lower antiproliferative effect on the cell line HCT 8 and the conjugate MP 264 and MP 265 showed lower cytotoxic effect than PTX...
