Huntingtonova choroba (HD) patří mezi neurodegenerativní onemocnění, od kterých se liší příčinou (expanze trinukleotidové repetice CAG v exonu 1 genu kódující protein huntingtin (Htt)) pouze v jednom genu (tzv. monogenní onemocnění). Ačkoliv je již od roku 1993 známá příčina nemoci, dosud není zcela známa patofyziologie HD a není nalezen lék. Pro lepší porozumění nemoci a pro možné preklinické testování se používají modely HD. Mezi nejčastěji používané modely patří hlodavci, hlavně myši, ale bylo důležité vytvořit i větší zvířecí modely, které by byly více podobné člověku. Proto bylo v roce 2009 vytvořeno na Akademii věd v Liběchově transgenní (TgHD) miniprase pomocí mikroinjikace lentivirového vektoru nesoucí N-terminální část lidského mutovaného HTT (mtHTT) s 124 CAG repeticemi v exonu 1. Tento model je životaschopný a v každé generaci se rodí určitá část Tg zvířat. V této práci jsem se věnovala biochemickým i behaviorálním změnám u tohoto modelu. Srovnávala jsem kontrolní (Wt) a transgenní (Tg) sourozence. Zjistila jsem, že biochemické změny se projevují hlavně zvýšenou přítomností fragmentů Htt ve varlatech a v mozku. V behaviorální části bylo mým hlavním úkolem zavedení metodiky pro další zkoumání behaviorálních změn u tohoto modelu, které zatím nebyly popsány. Byla testována zvířata F0, F1 a... Huntington's disease (HD) belongs to neurodegenerative disorders. It is a monogenic disease caused by trinucleotic CAG expansion in exon 1 of gene coding protein huntingtin. Even though the cause of HD is known since 1993, the pathophysiology and cure for HD reminds to be found. The animal models are being used for better understanding of HD. The most common animal models for HD are rodents, especially mice but it was also important to create large animal models, which will be more like human. Therefore, TgHD minipig was created in Academic of Science in Liběchov in 2009. This model was created by microinjection of lentiviral vector carrying N-terminal part of human HTT with 124 repetitive CAG in exon 1. This model is viable and in every generation, is part of the offspring transgenic. In this thesis, I specialized to biochemical and behavioral changes of this model. I compared transgenic and wild type siblings. I found that biochemical changes are manifested mostly by increased level of mtHtt fragments in testes and brain. In behavioral part of this thesis I established new methods for testing behavioral changes in this model.
