Apoptóza je jedním z mechanismů programované buněčné smrti, ke které dochází u nádorových buněk prsu po aplikaci taxanů. K její indukci může principielně docházet buď vnější nebo vnitřní dráhou. Diplomová práce má za cíl přispět k objasnění úlohy a mechanismů vnitřní mitochondriální dráhy indukce apoptózy po aplikaci taxanů (paclitaxel a SB-T-1216) na modelu buněčných linií karcinomu prsu SK-BR-3 (nefunkční protein p53, funkční kaspáza 3) a MCF-7 (funkční protein p53, nefunkční kaspáza 3). Konkrétně jsme sledovali vliv obou testovaných taxanů na mitochondriální membránový potenciál, hladinu ROS, expresi a lokalizaci proteinů regulujících vnitřní mitochondriální dráhu. U buněčné linie SK-BR-3 vedla aplikace obou taxanů k zhroucení mitochondriálního membránového potenciálu, avšak ne u buněčné linie MCF-7. Zjistili jsme, že se po aplikaci obou taxanů nemění exprese proteinu Bax a ani Smac/DIABLO u obou testovaných linií. U linie SK-BR-3 došlo na rozdíl od linie MCF-7 po aplikaci taxanů k poklesu exprese proteinu Bid. Po aplikaci taxanů nedošlo k translokaci proteinů Bax a Bid (tBid) z cytosolu na mitochondrie ani u jedné z testovaných linií. Nezjistili jsme také uvolňování proteinu Smac/DIABLO z mitochondrií do cytosolu. U buněčné linie SK-BR-3 jsme však pozorovali po aplikaci taxanů uvolňování... Apoptosis represents one of the cell death mechanisms which is realized after the application of taxanes in breast cancer cell lines. Apoptosis induction can be principally triggered either by outer or inner pathway. The aim of the diploma thesis is to contribute to the elucidation of role and mechanisms of the inner mitochondrial pathway of apoptosis induction after taxane application (paclitaxel and SB-T-1216) employing a model of breast carcinoma cell lines SK- BR-3 (nonfunctional p53, functional capase-3) and MCF-7 (functional p53, nonfunctional caspase-3). Specifically, we tested the effect of both employed taxanes on mitochondrial membrane potential, ROS level and the expression and localization of proteins regulating inner mitochondrial pathway. Taxane application resulted in mitochondrial membrane dissipation in SK-BR-3 cell line. However, this was not shown in MCF-7 cell line. We found no changes in Bax and Smac/DIABLO expression after taxane application in both tested cell lines. There was a decrease of Bid expression after taxane application in SK-BR-3 line, but not in MCF-7 line. Taxane application did not lead to the translocation of Bax and Bid (tBid) proteins from cytosol to mitochondria in both tested cell lines.
