Rhododendron tomentosum je stálezelený keř, který je typický svým obsahem sekundárních metabolitů, především esenciálního oleje s antimikrobiálními účinky. Diverzita endofytických hub v tomto druhu a jejich případná adaptace na růst v prostředí esenciálního oleje je velmi málo prozkoumána. Prvním cílem této práce tedy bylo zmapovat diverzitu endofytických hub kolonizujících listy druhu R. tomentosum na sedmi lokalitách v ČR a jedné v Estonsku. Z listů R. tomentosum jsem izolovala a pomocí srovnání úseků ITS1 a ITS2 rDNA se sekvencemi z GenBank a morfologických znaků určila 37 druhů endofytických hub. Dominantní byly druhy široce rozšířené a vyskytující se jako endofyté v zástupcích většiny rostlin. Druhým cílem mé práce bylo zjistit, zda esenciální olej z R. tomentosum má vliv na jeho endofytické houby. Předpokladem bylo, že kmeny endofytických hub získané z R. tomentosum budou růstu v prostředí esenciálního oleje přizpůsobené a na médiích s přidanými různými koncentracemi těchto chemických látek tedy porostou lépe v porovnání s kmeny stejných druhů získaných z jiných substrátů. U čtyř ze sedmi testovaných druhů rostly kmeny získané z R. tomentosum rychleji, ale i na médiu bez chemických sloučenin. Většinu kmenů (z R. tomentosum i z jiných substrátů) také výrazněji inhibovala až nejvyšší... Rhododendron tomentosum is an evergreen shrub with a high content of secondary metabolites, particularly essential oils with antimicrobial effects. Diversity of endophytic fungi in this species and their possible adaptation to growth in the essential oil environment is not much explored. Therefore, the first aim of this thesis was to reveal the diversity of endophytic fungi colonising leaves of R. tomentosum on seven localities in the Czech Republic and one in Estonia. I isolated and determined (using comparison of ITS1 and ITS2 rDNA with the sequences from GenBank and morphological signs) 37 species of endophytic fungi. Among them the ubiquitous species colonising the most of the plants as endophytes were dominant. The second aim of my thesis was to explore whether the essential oil from R. tomentosum influences its endophytic fungi. The hypothesis that the strains obtained from R. tomentosum would be adapted to growth in the environment of the essential oil was postulated. I supposed that they would grow better on mediums with different concentrations of these chemical compounds added, in comparison with strains of the same species obtained from different substrates.
