[cze][eng] 1 Abstrakt Traumatické míšní poranění (MP) je závažný stav s často velmi vážnými a trvalými následky. V současné době neexistuje žádná efektivní léčba. Po počátečním mechanickém poranění míchy se spouští kaskáda mnoha sekundárních dějů, které dále rozšiřují a zhoršují poškození. Tato předkládaná dizertační práce se zaměřuje na potenciální terapeutický vliv fotobiomodulace, přírodních protizánětlivých sloučenin epigallocatechin-3-galátu (EGCG), kurkuminu, jejich kombinace a čisté syntetické formy kurkuminu nazvané nanokurkumin na následky experimentálního MP u potkanů. Fotobiomodulace s využitím dvou synchronizovaných vlnových délek 808 a 905 nm vedla k rychlejšímu funkčnímu zotavení po MP, což bylo hodnoceno sérii lokomotorických testů a somatosenzorického plantar testu. Skupina léčená fotobiomodulací si vedla lépe ve všech sledovaných testech. Výsledky histopatologické analýzy ukázaly pozitivní vliv léčby na zachování šedé i bílé hmoty a naše výsledky rovněž naznačují upregulaci prospěšných M2 mikroglií/makrofágů po léčbě, což bylo hodnoceno imunohistochemickou a RT-qPCR analýzou. Výsledky ukazují, že fotobiomodulace je nadějnou neinvazivní metodou pro zlepšení motorických funkcí a ochranu míšní tkáně po MP. EGCG a kurkumin jsou přírodní sloučeniny známé v čínské medicíně již po staletí. Ve druhé části...1 Abstract Traumatic spinal cord injury (SCI) is severe condition with devastating long- lasting consequences. There is still no available treatment. After initialing mechanical trauma, there is a huge cascade of secondary reactions which amplifies the damage. This thesis is focused on potential therapeutic effect of photobiomodulation, natural anti- inflammatory compounds epigallocatechin-3-gallate (EGCG), curcumin, their combination and extrapure synthetic form of curcumin called "nanocurcumin" on impacts of experimental model of SCI in rats. Photobiomodulation using combination of two synchronized wavelengths 808 and 905 nm improved functional recovery after SCI evaluated by battery of locomotor tests and somatosensory plantar test. The group treated by photobiomodulation obtain better results in all tests. The histopathological analysis showed a positive effect on white and gray matter sparing and our data suggests an upregulation of M2 microglia/macrophages in photobiomodulation treated rats assessed by immunohistochemical and RT-qPCR analysis. Our results demonstrated that the photobiomodulation is a promising non-invasive therapy for improving functional recovery and tissue sparing after SCI.
