Souhrn Úvod a cíl: Neuropeptidy tvoří rozsáhlou skupinu biologicky aktivních látek a jejich účinky závisí na aktivitě a lokalizaci specifických receptorů. Dosud není zcela objasněn mechanismus regulačních/modulačních účinků neuropeptidů a tyto mezery v našich znalostech pak ztěžují možnost jejich terapeutického ovlivnění. Naší hypotézou bylo, že podobně jako byly popsány účinky galaninergního systému na kardiovaskulární systém, může být galaninergní systém jedním z významných modulátorů hypotalamo-hypofýzo-adrenální osy a může proto modulovat stresovou odpověď či patofyziologii vybraných neurologických onemocnění. Cílem naší práce bylo zjistit přítomnost a expresi galaninových (GalR) a CRH (CRH-R) receptorů za bazálních podmínkách a při stresu v adenohypofýze a hypofýze a za patologických podmínek v míše na myším modelu roztroušené sklerózy, experimentální autoimunitní encefalomyelitidy (EAE) a určit buněčnou lokalizaci GalR1 (oligodendrocyty, mikroglie, astrocyty, ependymální buňky a endotelové buňky v kapilárách). Výsledky: Exprese všech testovaných galaninergních peptidů byla stanovena v adenohypofýze. Bylo zjištěno, že exprese mRNA GalR2 v adenohypofýze za bazálních podmínek byla mnohem vyšší než exprese mRNA GalR1 a GalR3. Zatímco akutní stres nevyvolal v adenohypofýze žádné změny v expresi GalR2,... Introduction and Aim: Neuropeptides are widely distributed group of biologically active substances and their effects depend on their activity and localization of specific receptors. The mechanism of the regulatory / modulatory effects of neuropeptides has not been fully elucidated yet, and these gaps in our knowledge make it difficult for them to influence them therapeutically. Our hypothesis was that similar to the effects of the galaninergic system on the cardiovascular system, the galaninergic system may be one of the important modulators of the hypothalamic-pituitary-adrenal axis and may therefore modulate the stress response or pathophysiology of selected neurological diseases. The aim of our study was to investigate presence and expression of CRH and Galanin receptors in the basal conditions and in stress in adenohypohysis and in hypophysis and in pathological conditions - in the spinal cords in the murine model of multiple sclerosis, experimental autoimmune encephalomyelitis (EAE) and determine GalR1 cellular localization (oligodendrocytes, microglia, astrocytes, ependymal cells, and endothelial cells in the capillaries).
