Úvod/Cíle: Trombofílie, tedy zvýšená dispozice ke vzniku žilní a arteriální trombózy představuje komplexní onemocnění zapříčiněné mimo jiné i poruchami krevních destiček a endotelu. Cirkulující endotelové buňky (CEC) a endotelové progenitorové buňky (EPC) jsou popisovány jako markery poškození a dysfunkce endotelu, respektive jako markery jeho obnovy u mnoha nemocí, včetně trombotických komplikací. Jejich význam u pacientů se známou trombofílií však dosud ještě nebyl zkoumán. CEC i EPC představují extrémně vzácné buněčné populace vyskytující se v periferní krvi. Proto k jejich identifikaci a kvantifikaci je použití výhradně standardizovaných a citlivých metod zcela nezbytné. Cílem práce bylo identifikovat a kvantifikovat CEC a EPC v periferní krvi pacientů s vrozenými trombofiliemi a zhodnotit jejich význam jako markerů aktivity endotelu a krevních destiček v souvislosti s rizikem vzniku či rekurence trombózy. Metody: Analýza počtu CEC a EPC v periferní krvi pacientů s trombofiliemi s nebo bez historie trombózy a pacientů s akutní trombózou byla provedena metodou vícebarevné průtokové cytometrie. Referenční hodnoty CEC a EPC byly stanoveny na skupině zdravých kontrol. Background/Aims: Thrombophilia, ie an increased predisposition to venous and arterial thrombosis, is a complex disease caused by disorders of platelets and endothelial, among others. Circulating endothelial cells (CEC) and endothelial progenitor cells (EPC) have been described as markers of endothelial damage and dysfunction, respectively as markers of its recovery in many diseases, including thrombotic complications. However, their significance in patients with known thrombophilia has not yet been investigated. Both CEC and EPC represent extremely rare cell populations found in peripheral blood. Therefore, it is essential to use exclusively standardized and sensitive methods for their identification and quantification. The aim of the study was to identify and quantify CEC and EPC in the peripheral blood of patients with congenital thrombophilia and to evaluate their importance as markers of endothelial and platelet activity in context with the risk of thrombosis occuring and recurrence.
