Metabolické transkripční represory jsou proteiny, které regulují transkripci různých genů bakteriálního metabolismu. Typicky obsahují dvě domény: N-koncovou doménu vázající DNA (DBD, z angl. DNA-binding domain) a C-koncovou doménu vázající molekulu efektoru. Po vazbě efektoru (nejčastěji molekuly metabolitu) protein změní svou konformaci, čímž dojde ke změně jeho afinity k DNA operátoru a následné modulaci transkripce genů kódujících proteiny dané metabolické dráhy. LutR patří do rodiny bakteriálních transkripčních regulátorů GntR. V divokém kmeni Bacillus subtilis RO-NN-1 je LutR zodpovědný za represi transkripce genů potřebných pro využití laktátu, zatímco funkce LutR z laboratorních kmenů B. subtilis PY79 a 168 se liší. Díky mutaci je produkovaný protein o prvních 21 aminokyselin kratší, což výrazně ovlivňuje jeho vazebnou specificitu při rozpoznávání sekvence DNA. Tato varianta LutR pak funguje jako globální regulátor a reguluje velký počet genů, jejichž transkripce je spjata s přechodem z fáze exponenciálního růstu bakteriální populace do fáze stacionární. Znalost 3D struktury komplexu LutR s DNA by pomohla objasnit přesný mechanismus, jakým delece v DBD LutR mění její vazebnou specificitu. V rámci této práce byla heterologní expresí v Escherichia coli připravena DBD LutR z divokých kmenů B.... Metabolic transcriptional repressors are proteins controlling transcription of specific genes involved in bacterial metabolism. These proteins typically consist of two domains: N-terminal DNA-binding domain (DBD) and C-terminal effector-binding domain. When an effector (usually a metabolite molecule) binds to the protein, the conformation of the protein is changed. This causes a change in affinity to its DNA operator and that subsequently modulates the transcription of genes of the specific metabolic pathway. LutR belongs to the GntR family of bacterial transcriptional regulators. In undomesticated strain RO-NN-1 of Bacillus subtilis, LutR regulates transcription of genes required for L-lactate utilization. Interestingly, LutR from laboratory strains PY79 and 168 has a different function. Due to a mutation, it lacks the first 21 amino acids and this alters its DNA recognition specificity. This LutR variant acts as a global regulator and regulates many genes associated with transition from exponential growth to stationary phase of bacterial population. Knowledge of 3D structure of LutR with DNA could elucidate the impact of this short deletion in LutR DBD on the mechanism of DNA recognition.
