Práce je zaměřena na možnosti využití techniky Ramanovy spektroskopie kapkově nanášených povlaků (DCDR) v oblasti studia cukrů. DCDR spektroskopie nabízí snížení detekčního prahu pomocí efektu kávového kroužku a minimalizaci signálu vody při zachování podobnosti se spektrem získaným z roztoku. Podobnosti je dosaženo díky skelné fázi, v níž mají molekuly náhodnou orientaci a pravděpodobně zachovanou první solvatační obálku. U malých molekul je však situace komplikována jak nízkou účinností efektu kávového kroužku, tak tendencí ke krystalizaci. V práci jsme studovali glukózu, galaktózu, laktózu, maltózu, trehalózu a cyklodextrin, tak abychom pokryli základní strukturní rozmanitost cukrů - od monosacharidů, disacharidů až po oligosacharidy. Podařilo se úspěšně nalézt koncentrační rozmezí pro DCDR spektroskopii mono a disacharidů (ca. 1-30 mM), při zachování skelné fáze depozitu, a ukázat na komplikace spojené s krystalizací u polysacharidů. Získaná DCDR spektra jsme porovnali se spektry polykrystalických vzorků, od nichž se jasně odlišovala. Následně jsme proměřili koncentrační závislosti, až do nasycených roztoků, jejichž Ramanova spektra jsme analyzovali pomocí faktorové analýzy ve snaze podchytit spektrální změny indukované koncentrací, a tedy narušením solvatačních obálek. Byly však nalezeny... The work is focused on the usage of drop-coating Raman (DCDR) spectroscopy in the study of carbohydrates. DCDR spectroscopy offers a lowering of the detection limits by the coffee-ring effect and minimization of the water signal while keeping similarity to the spectrum obtained from the solution. The similarity is achieved due to the glassy phase, in which molecules have a random orientation and probably retain the first solvation envelope. However, for small molecules, the situation is complicated by both the low efficiency of the coffee-ring effect and the tendency to crystallize. In this work, we studied glucose, galactose, lactose, maltose, trehalose and cyclodextrin to cover the basic structural diversity of sugars - from monosaccharides, disaccharides to oligosaccharides. It was successfully found the concentration range for DCDR spectroscopy of mono and disaccharides (ca. 1-30 mM), while maintaining the glassy phase of the deposit, and to point out the complications associated with the crystallization of polysaccharides.
