[cze][eng] 4 Abstrakt Kvasinky, ač se jedná o jednobuněčné mikroorganismy, v přirozených podmínkách vytvářejí složité mnohobuněčné útvary, jako jsou například biofilmy a kolonie. V rámci těchto útvarů následně probíhají děje, jako buněčná diferenciace, specializace jednotlivých buněčných populací a buněčná signalizace, typické pro mnohobuněčné organismy. V literárním úvodu této práce je shrnut přehled současných znalostí o vývoji biofilmů a kolonií, a to především druhu Saccharomyces cerevisiae, jako modelovém organismu, a některých vybraných regulací, které jsou důležité pro tvorbu mnohobuněčných struktur. Ve výsledkové části se zaměřuji na dva směry výzkumu. První z nich je směřován na mechanizmy podílející se na tvorbě mnohobuněčných struktur. Při studiu formování SLI biofilmů (biofilmů na rozhraní pevný povrch/kapalina) jsme zdokumentovali antagonistickou roli regulátorů CYC8 a TUP1 na jejich utváření a také jsme popsali vliv přítomnosti glukózy na vývoj a stabilitu SLI biofilmů kmene BR-F. Při této příležitosti jsme vyvinuli zobrazovací metodu, díky které jsme schopni za fyziologických podmínek připravit a pomocí mikroskopie pozorovat vnitřní strukturu (svislý průřez) SLI biofilmů i nárůstů neadherovaných buněk. Dále jsme popsali principy distribuce buněk při růstu obřích kolonií a zavedli model distribuce...5 Abstract Although yeasts are unicellular microorganisms, they form complex multicellular formations such as biofilms and colonies under natural conditions. Within these structures, processes such as cell differentiation, specialization by particular cell populations and cell signalling, which are typical of multicellular organisms, take place. The literature introduction to this thesis summarizes current knowledge regarding the development of biofilms and colonies, in particular those of the model organism, Saccharomyces cerevisiae, and some selected regulations that are important for the formation of multicellular structures. In the results section, I focus on two lines of research. The first is directed towards mechanisms, involved in the formation of multicellular structures. In studying the formation of SLI biofilms (biofilms at the solid/liquid interface), we have documented the antagonistic role of the regulators CYC8 and TUP1 in their formation and have also described the effect of the presence of glucose on the development and stability of SLI biofilms of strain BR-F.
