[cze][eng] V této bakalářské práci byly připraveny a charakterizovány hydrogely na bázi 2-hydroxyethylmethakrylátu a kyseliny methakrylové (pHEMA-MA). Hydrogely by mohly sloužit jako testovací médium při vývoji nanovlákenných polylaktidových (PLA) systémů pro lokální aplikaci paklitaxelu (PTX). Toto médium bylo studováno ve snaze vytvořit podmínky, které lépe odpovídají skutečnému prostředí lidského těla než běžně používaná kapalná média. Lokální aplikace cytostatik u léčby pevných nádorů má řadu výhod. PTX je představitelem hydrofobního protinádorového léčiva s vážnými vedlejšími účinky. Lokální aplikací tohoto léčiva pomocí PLA nanovlákenných nosičů by bylo možné vedlejší účinky značně omezit a při tom zvýšit jeho koncentraci a dobu působení v cílových nádorových tkáních. Dále byl zkoumán vliv přídavku polyethylenglykolu (PEG) o molekulové hmotnosti 20 kDa a β-cyklodextrinu (CD) do struktury nanovláken pro optimalizaci uvolňování inkorporovaného léčiva. Kinetika uvolňování PTX z PLA nanovláken do pHEMA-MA hydrogelů byla sledována analytickou metodou HPLC s UV detekcí. Experiment byl také proveden za použití vodného roztoku PBS jako uvolňovacího média pro porovnání profilů uvolňování PTX. Za použití metody HPLC s UV detekcí byla testována také sorpce PTX do pHEMA a pHEMA-MA hydrogelů o různém obsahu MA z...In present bachelor thesis, hydrogels based on 2-hydroxyethyl methacrylate and methacrylic acid (pHEMA-MA) were prepared and characterized. Hydrogels could serve as a testing medium in the development of nanofiber polylactide (PLA) systems for local application of paclitaxel (PTX). This medium has been studied in an effort to create conditions that better correspond to the actual environment of a human body than commonly used liquid media. Local application of cytostatics in the treatment of solid tumors has a number of advantages. PTX represents a hydrophobic anticancer drug with serious side effects. Local distribution of presented drug using nanofiber carriers could greatly reduce the adverse side effects while increasing its concentration and duration of effect in target tumor tissues. The effect of the addition of polyethylene glycol (PEG) with a molecular weight of 20 kDa and β-cyclodextrin (CD) to the nanofiber structure was further investigated to optimize the release of the incorporated drug. The release kinetics of PTX from PLA nanofibers into pHEMA-MA hydrogels were studied using analytical method HPLC with UV detection.
