Dlaždicové karcinomy hlavy a krku jsou i přes pokroky medicíny v posledních desetiletích výzvou na poli onkologické léčby. Studium molekulární biologie umožňuje blíže charakterizovat vlastnosti nádorů a předpovědět prognózu pro postižené pacienty. Současně je vyvíjeno a klinicky testováno několik léků z kategorie cílené terapie. Experimentální práce spočívala jak v in vitro, tak v in situ pokusech prováděných na základě spolupráce mezi několika pracovišti 1.LF UK, Akademie věd ČR, ÚHKT a Fakultou veterinární medicíny Ludwig-Maximilianovy univerzity. Galektinu-1 je významným induktorem vzniku myofibroblastů/nádorově asociovaných fibroblastů. Tyto fibroblasty jsou, díky své schopnosti indukovat invazivní chování nádorových buněk, považovány za nositele špatné prognózy onemocnění. Galektin-9 naopak není v karcinomu exprimován a v případě dysplastické tkáně, prokázané například aberantní expresi keratinu 14 a 19, dochází též k aberantní expresi gal-9. Kromě využití galektinů jako prognostických znaků je studován též jejich význam terapeutický. Prezentovaná práce s mutovanými variantami galektinu-2 ukázala možnosti ovlivnění jak farmakodynamiky, tak farmakokinetiky upravených galektinů. Výsledky ukázaly prodloužení biologického rozpadu PEGylovaného galektinu na úkor jeho schopnosti tlumit proliferaci... Head and neck squamous cell carcinomas are still challenging despite progress in the oncological treatment. Study of the molecular biology allows to deeply characterize tumor properties and to predict the prognosis for affected patients. Nowadays there are many drugs clinically tested in the group of targeted therapy medicine Experimental work comprised both in vitro and in situ assays, being performed thanks to the collaboration between a number of departments of the 1st Faculty of Medicine of the Charles University in Prague, Academy of Sciences of the Czech Republic, Institute of Hematology and Blood Transfusion and Faculty of Veterinary Medicine of the Ludwig-Maxmillian University Munich. Galectin-1 is important inductor of the myofibroblasts/cancer associated fibroblasts. These fibroblasts are regarded as negative prognostic markers thanks to their capability of invasive cancer cells induction. On the other hand, Galectin-9 is not present in the carcinoma and in the case of dysplasia, its expression indicate aberrant features together with aberrant expression of keratin 14 and 19.
