[cze][eng] Souhrn V této dizertační práci byly studovány: 1) potenciálně kardioprotektivní účinky deferipronu na modelu daunorubicinem navozené chronické kardiotoxicity u králíků, 2) úloha apoptotické buněčné smrti při rozvoji antracyklinové kardiotoxicity, 3) kardioprotektivní účinky dexrazoxanu vůči chronické antracyklinové kardiotoxicitě se zaměřením na ochranu kardiomyocytů před programovanou buněčnou smrtí a oxidačním stresem a 4) časový sled změn vybraných parametrů v průběhu rozvoje chronické antracyklinové kardiotoxicity. Na leukemické buněčné linii bylo ukázáno, že deferipron (1-300 µmol/l) nesnižuje protinádorový účinek daunorubicinu, naopak při vyšších koncentracích deferipronu byla pozorována sumace antiproliferativních účinků obou látek. Ve vlastní kardioprotektivní studii však podání deferipronu nezabránilo daunorubicinem navozené mortalitě, srdeční dysfunkci, morfologickým změnám v myokardu, nárůstu srdečního troponinu T a stejně tak peroxidaci lipidů v myokardu. Tyto výsledky kontrastují s předešlými pozitivními závěry in vitro studií, a tudíž značně mění dosavadní pohled na deferipron jako na potenciální kardioprotektivum vůči antracyklinové kardiotoxicitě. Získané výsledky společně s předchozími nálezy navíc naznačují, že role železa a jeho chelatace při antracyklinové kardiotoxicitě je mnohem...In this Ph.D. thesis, following aims were addressed: 1) potentially cardioprotective effects of deferiprone on the model of daunorubicin-induced chronic cardiotoxicity in rabbits, 2) the role of apoptotic cell death in the development of anthracycline cardiotoxicity, 3) cardioprotective effects of dexrazoxane against chronic anthracycline cardiotoxicity with a focus on rescue of cardiac myocytes from programmed cell death and oxidative stress, and 4) staging of myocardial changes in the time-course of chronic anthracycline cardiotoxicity development. First, using the leukemic cell line, deferiprone (1-300 µmol/L) was shown not to blunt the antiproliferative effect of daunorubicin. Instead, at higher concentrations of deferiprone, the augmentation of antiproliferative actions of both agents was observed. However, in the cardioprotective study deferiprone failed to afford significant protection against daunorubicin-induced mortality, cardiac dysfunction, morphological cardiac deteriorations, plasma cardiac troponin T rise as well as myocardial lipoperoxidation. This finding contrasted with previous positive outcomes of in vitro studies.
