RNDr. Lenka SKÁLOVÁ, Ph.D. Název disertační práce METABOLISMUS A ÚČINKY NOVÝCH ANTHELMINTIK U HELMINTŮ A JEJICH HOSTITELŮ Onemocnění způsobené helminty, tzv. helmintózy, jsou jedním z největších problémů ve veterinární i humánní medicíně. Tyto nákazy se léčí anthelmintiky, avšak jejich účinnost není často dostatečná kvůli rezistenci helmintů vůči běžným anthelmintikům. Celosvětové rozšíření resistence helmintů vedlo k vývoji nových léčiv, která mají odlišný mechanismus účinku než léčiva dosud používaná. Monepantel (MOP) patří do nové třídy anthelmintik, známé jako amino- acetonitrilové deriváty (AAD) s účinkem na nikotin-acetylcholinové receptory. MOP byl schválen ve veterinární praxi v České republice v roce 2011 pro terapii gastrointestinálních onemocnění, která jsou způsobená hlísticemi. Komerčně je vyráběn pod názvem Zolvix® od firmy Novartis AG. Cílem předložené disertační práce bylo studovat metabolismus a účinky MOP a jeho derivátů za účelem přispět ke globálnímu výzkumu na tomto poli. V experimentech byly jako zástupci hlístic použity Vlasovky slézové (Haemonchus contortus, citlivý kmen ISE a rezistentní kmen WR) a jejich hostitelé ovce (Ovis spp). V... RNDr. Lenka SKÁLOVÁ, Ph.D. Title of Doctoral Thesis METABOLISM AND EFFECTS OF NEW ANTHELMINTIC DRUG IN HELMINTHS AND ITS HOSTS Diseases caused by helminths, called helminthoses, are one of the most serious problems in veterinary and human medicine. These diseases are treated by anthelmintics but their effectiveness is often not sufficient due to resistance of helminths against common anthelmintics. The global anthelmintic resistance encouraged the developing of new drugs with a different mode of action. Monepantel (MOP) belongs to a new class of anthelmintic drugs known as amino- acetonitrile derivatives (AADs) that act on the nicotinic acetylcholine receptors. In 2011, MOP was approved for the treatment of gastrointestinal diseases caused by nematodes in the veterinary practice in the Czech Republic. It is produced commercially under the trade name Zolvix® (Novartis AG).
