Bakterie Bordetella pertussis je lidský patogen a původce černého kašle, vážné respirační nemoci, jejíž výskyt se v průmyslově rozvinutých zemích v posledních desetiletích zvýšil. Jedním z dosud málo prostudovaných faktorů virulence B. pertussis je sekreční systém typu 3 (T3SS) zajišťující transport efektorových proteinů produkovaných gramnegativními bakteriemi do hostitelských buněk. V této diplomové práci byly studovány faktory ovlivňující aktivitu T3SS in vitro. Bylo zjištěno, že T3SS laboratorního kmene Tohama I lze aktivovat kontaktem buněk s biologicky aktivními termolabilními složkami krve (např. komplement). Naopak u nedávno izolovaného kmene B1917 je T3SS aktivní i bez indukce krví. Indukce T3SS in vitro byla dále studována u nově připraveného kmene bakterie B. bronchiseptica RB50 postrádajícího gen BB0302 kódující regulátor transkripce rodiny GntR (homolog BP0209 bakterie B. pertussis). Delece genu BB0302 však na aktivitu T3SS neměla vliv. Odolnost vůči baktericidnímu působení krevního séra je důležitým faktorem, který hraje roli v patogenezi B pertussis. Bylo zjištěno, že české klinické izoláty (2008-2015) jsou vůči séru in vitro výrazně odolnější než původní vakcinační kmeny (1954-1965). Tento jev pravděpodobně souvisí s postupnou adaptací globální populace B. pertussis na lidského... Bordetella pertussis is a Gram-negative strictly human pathogen and the major causative agent of whooping cough or pertussis. The incidence of this highly contagious respiratory disease in developed countries has increased in the last decades. One of the less characterized virulence factors of B. pertussis is the type three secretion system (TTSS) which is responsible for the secretion of the effector proteins into host eukaryotic cells. This diploma thesis sheds light onto factors influencing TTSS in vitro activity. Although TTSS of laboratory strain Tohama I was induced by biologically active compounds present in blood (e. g. complement proteins), TTSS of recent clinical isolate B1917 seems to be induced permanently. Furthermore, BB0302 encoding a GntR family transcription regulator in B. bronchiseptica RB50 (homologous to BP0209 of Tohama I) was studied, however, the deletion of this gene did not affect the TTSS functionality. Serum resistance is a factor that plays a key role in the pathogenesis of B pertussis. We show that Czech recent isolates (2008-2015) are significantly more resistant to serum killing in vitro than the original vaccine strains (1954-1965).
