Dermatofyty jsou skupina hub, z nichž některé mohou díky své schopnosti rozkládat keratinizovanou tkáň způsobovat onemocnění kůže u lidí i zvířat. Mezi zástupce této skupiny patří také kmeny komplexu Trichophyton benhamiae, známí původci dermatofytóz zejména drobných hlodavců a králíků. V poslední dekádě se jedna ze čtyř populací tohoto komplexu epidemicky šíří Evropou mezi morčaty a jejich chovateli. Aby mohla být zodpovězena otázka, co stojí za úspěšným šířením této populace, byla zkoumána genová exprese a produkce volatilních organických látek epidemické a neepidemických populací T. benhamiae. Genová exprese tří kmenů od každé populace byla studována při růstu v tekutém médiu a na ex vivo myších kožních modelech připravených podle nově optimalizovaného protokolu. Pro analýzu genové exprese byly zvoleny metody RNAseq a RT-qPCR. Na základě literatury a výsledků předběžné analýzy RNAseq bylo vybráno několik genů, pro které byly navrženy specifické primery. Spektra produkovaných volatilních organických látek stejných kmenů rostoucích na ovčí vlně ve vialkách byla analyzována metodou GC-MS. Zatímco neepidemické populace se v genové expresi a produkci volatilních organických látek nelišily, epidemická populace se ukázala být odlišná od ostatních zástupců studovaných skupin. Podle získaných dat... Dermatophytes are a group of fungi, some of which can cause skin diseases in humans and animals due to their ability to degrade keratinized tissue. Representatives of this group also include strains from the Trichophyton benhamiae complex, known to cause dermatophytosis especially of small rodents and rabbits. In the last decade, one of four populations of this complex has spread epidemically across Europe among guinea pigs and their breeders. To answer the question what stands behind the successful spread of this population, the gene expression and production of volatile organic compounds of epidemic and non-epidemic populations of T. benhamiae was investigated. Gene expression of three strains from each population was studied during growth in liquid medium and on ex vivo mouse skin models prepared according to a newly optimized protocol. RNAseq and RT-qPCR methods were chosen for the gene expression analysis. Based on the literature and the results of RNAseq preliminary analysis, several genes were selected for which specific primers were designed. The spectra of the produced volatile organic compounds of the same strains growing on sheep wool in vials were analyzed by GC-MS.
