Komplex exocyst je osmi-proteinový komplex známý též jako Sec6/8 a jeho kompozice je evolučně vysoce konzervovaná napříč organismy. Tento komplex se účastní váčkového transportu jakožto součást poutacího mechanismu na specifické místo plazmatické membrány. V genomu Arabidopsis thaliana se jeho podjednotka EXO70 vyskytuje ve 23 kopiích tohoto genu. V práci byly studované paralogy EXO70H7 a EXO70H8, u kterých podle dosud dostupných informacích vzniklo podezření na důležitost ve vývoji kořene. U rostlin mutantních v těchto genech se během práce nepodařilo identifikovat žádný výrazný fenotypový projev. Pomocí experimentů za stresových podmínek byly testovány další projevy mutací v těchto genech. Většina těchto experimentů neidentifikovala žádné odchylky. Pouze při klíčení semen za stresových podmínek bylo zjištěno, že dochází k signifikantně horšímu klíčení mutantních semen exo70H7 na médiích obsahujících sorbitol, což poukazuje na horší obranu proti osmotickému stresu. Signifikantní zhoršení v klíčení semen exo70H8 na médiu s nadbytkem NaCl naznačuje větší náchylnost k nadbytečnému příjmu iontů sodíku. Analýza buněčné lokalizace pomocí vytvořených konstruktů s GFP přinesla poznání lokalizace těchto proteinů. EXO70H7 se lokalizuje v cytoplazmě a v jádře s negativně kontrastním jadérkem a lokalizace... Complex Exocyst consists of eight proteins and it is known as a Sec6/8. Its composition is evolutionarily highly conserved amongst all the species. This complex is involved in vesicle trafficking as a part of attaching mechanism to a specific place on the plasma membrane. EXO70 subunit has been found in 23 copies in Arabidopsis thaliana genome. In this study we have been examine paraloges EXO70H7 and EXO70H8. There have been suggestions that these genes are important in development of roots according to the previous studies. We have not been able to identify any significant phenotype within the mutant plants in these genes. There has been studied other mutant appearance during the stress experiments. Most of these experiments did not identify any divergence. Only experiments with germination during stress conditions revealed significantly worse germination of exo70H7 mutant seeds on the medium containing sorbitol. This suggests that mutant seeds have a worse protection against osmotic stress. Significantly worse germination of exo70H8 seeds on the medium with excess NaCl indicates that these seeds incriminate to higher absorption of sodium ions.
