Nelegální obchod s volně žijícími živočichy a rostlinami je jedním z nejlukrativnějších zločinů na světě. Hnací silou této nekalé praktiky ohrožující mnoho druhů na přežití je z velké časti stále se zvyšující poptávka po surovinách k přípravě tradiční čínské medicíny (TCM). Její léčebné přípravky přitom zahrnují mimo jiné i organizmy, s nimiž je obchod zakázán, nebo regulován podle úmluvy CITES (the Convention on International Trade in Endangered Species of Wild Fauna and Flora). Tato mezinárodní dohoda jasně definuje pravidla obchodu s volně žijícími druhy i výrobky z nich. Jejich porušení se trestá na základě legislativy konkrétního státu. K zefektivnění potírání tohoto typu kriminality je zapotřebí snadno standardizovatelných a přesných technik druhové identifikace. Ve své práci uvádím souhrn a zhodnocení metod DNA identifikace založených na principu hybridizace, polymerázové řetězové reakce nebo sekvenaci, a to i pro vzorky s degradovanou DNA, nebo komplexní vzorky s obsahem směsi různých druhů. DNA barcoding se zdá být optimálním nástrojem k detekci zdrojových organizmů čínské medicíny. Text je doplněn o výčet nejčastěji zadržovaných exemplářů CITES organizmů využívaných pro účely TCM s fotografiemi artefaktů zabavených Českou inspekcí životního prostředí. Tato práce by tak mohla sloužit... The illegal trade of wildlife animal and plant species is one of the most lucrative crimes in the world. The driving force behind this unfair practice of threatening many species for survival is the largely increasing demand for raw materials used to prepare traditional Chinese medicine (TCM). Its medicinal preparations include, among others, organisms with which trade is prohibited or regulated under the CITES convention (the Convention on International Trade in Endangered Species of Wild Fauna and Flora). This international agreement clearly defines the rules of trade of wild species and products thereof, the violation of which is punishable on the basis of the legislation of a particular country. Easily standardized and accurate species identification techniques are needed to make more effective the prosecution of this type of crime. In my bachelor's thesis I am representing a summary and evaluation of DNA identification methods, which are based on the principle of hybridization, polymerase chain reaction or sequencing, including samples with degraded DNA or complex samples containing a mixture of different species.
