Během těhotenství se mateřský organismus musí vypořádat s přítomností cizorodých paternálních antigenů, umožnit a podporovat v sobě vývoj semialogenního plodu, a přitom zachovat svou obranyschopnost vůči patogenům. Tento stav umožňuje dynamická rovnováha komplexních imuno-endokrinních interakcí. Na tolerogenním nastavení mateřského organismu v těhotenství se podílí množství buněk se specifickými vlastnostmi. Velmi významnou je populace T regulačních lymfocytů (Treg). Množství Treg v průběhu těhotenství fyziologicky stoupá a jejich pokles je spojen s porodem termínovým i předčasným. Předčasný porod představuje závažný socioekonomický problém, který se celosvětově týká přibližně 10 % porodů. V případě zvýšeného rizika předčasného porodu se v klinické praxi s úspěchem využívá podávání progesteronu. Jednou z hypotéz je, že progesteron indukuje zvýšení populace Treg a touto cestou oddaluje předčasný porod. Cílem diplomové práce bylo sledovat vliv progesteronové terapie in vivo na množství a funkční vlastnosti CD4+ CD25+ FoxP3+ Helios+/- Treg a ověřit, zda dlouhodobé vaginální podávání progesteronu pozitivně koreluje se zvýšením počtu Treg. Dalším cílem bylo zjistit, zda progesteron podporuje vznik Treg a tvorbu imunoregulačních cytokinů in vitro. Hlavní použitou metodou je průtoková cytometrie.... During pregnancy, the maternal organism has to cope with the presence of foreign paternal antigens, allow and support the development of a semialogenic fetus, while maintaining its defense against pathogens. This condition is achieved by a dynamic balance of complex immuno-endocrine interactions. A number of cells with specific properties contributes to the tolerogenic setting during pregnancy. Very important role plays the population of T regulatory lymphocytes (Treg). Treg levels increase physiologically during pregnancy and their decline is associated with term and preterm labor. Premature birth is a serious socio-economic problem, affecting around 10 % of births worldwide. In the case of increased risk of premature labor, progesterone administration has been successfully used in clinical practice. One hypothesis says that progesterone induces an increase in Treg population and delays premature labor in this way. The aim of the thesis was to monitor the effect of progesterone therapy in vivo on the amount and functional properties of CD4+ CD25+ FoxP3+ Helios+/- Treg and to verify whether long-term vaginal administration of progesterone positively correlates with an increase in Treg counts.
