Rakovina patří mezi nejzávažnější problémy, se kterými se potýká moderní medicína. V posledních letech došlo k velkému rozvoji nanotechnologií a jejich použití v medicíně. Cílem je zefektivnit podávání léčiv a přispět tak k léčbě nádorových onemocnění. Uložení látky do nanočástice může být řešením při nízké stabilitě léčiva a/nebo může přispět k eliminaci nežádoucích vedlejších účinků. Nanočástice apoferritin, která byla sledována v této bakalářské práci, je forma běžně se vyskytujícího proteinu ferritinu. V jeho struktuře se nachází dutina, do které je možné vložit požadované léčivo. Jeho chemické vlastnosti (vysoká stabilita při extrémních teplotách a široké škále pH, snadná manipulace pomocí změny pH) a také biokompatibilita z něj dělají potenciálně vhodný transportér. V předkládané bakalářské práci byla studována schopnost apoferritinu inkorporovat do své struktury protinádorové léčivo cabozantinib. Jedná se o tyrosinkinázový inhibitor 'používaný pro léčbu karcinomu štítné žlázy, ledvinových buněk a hepatocelulárního karcinomu. Dále byl sledován vliv konečné hodnoty pH na vznik komplexu apoferritinu s cabozantinibem a také stabilita tohoto komplexu. Ze získaných výsledků vyplývá, že apoferritin může enkapsulovat cabozantinib do své vnitřní struktury, přičemž konečná hodnota pH na tento proces... Cancer is one of the most serious problems, which modern medicine faces. In recent years, nanotechnologies and their use in medicine, has developed greatly. The aim is to make drug administration more effective and help to improve treatment of cancer illnesses. Incorporation of chemical substance into a nanoparticle can solve the problem with low stability of the drug, and/or it help to eliminate side effects. Nanoparticle apoferritin, which was studied in this thesis, is a form of commonly occurring protein ferritin. Its structure contains cavity, that can be used for incorporation of drug. Its chemical structure (high temperature stability and stability at wide pH range, easy manipulation by changing pH) and its biocompatibility makes apoferritin a potentionally suitable transporter. Presented thesis studied apoferritin's ability to incorporate anticancer drug cabozantinib into its structure. Cabozantinib is tyrosine kinase inhibitor which is used for treatment of thyroid cancer, renal cell carcinoma and hepatocellular carcinoma. The effect of final pH to the formation of the complex of apoferritin with cabozantinib, and stability of this complex was also studied in this thesis.
