Inhibitory tyrosinkinas (TKI) jsou malé organické molekuly, které se používají jako léčiva při cílené protinádorové terapii. V nádorových buňkách inhibují vybrané aktivované receptorové tyrosinkinasy a brání tak proliferaci, růstu a metastázám nádoru, a angiogenesi v nádorové tkáni. Lenvatinib a vandetanib jsou inhibitory tyrosinkinas, které se používají pro léčbu karcinomu štítné žlázy. Předkládaná diplomová práce rozšiřuje poznání cest využitelných ke zvýšení účinnosti daných protinádorových léčiv. Jedno ze studovaných protinádorových léčiv - lenvatinib - jsme se pokusili připravit ve formě nanočástic. Jako transportní částice jsme využili protein apoferritin a liposomy. Počítačové modelování interakce lenvatinibu se strukturou apoferritinu a model jeho enkapsulace signalizovalo, že toto léčivo není vhodné pro přípravu apoferritinových nanočástic. A to vzhledem k tomu, že se při jejich přípravě vyskytuje převážně v neutrální formě. Tento předpoklad potvrdily i experimenty, ve kterých se částice lenvatinibu v apoferritinu skutečně nepodařilo připravit. Použitý počítačový model nám tak může do budoucnosti posloužit pro "screening" konstrukce protinádorových léčiv ve formě nanočástic ještě před samotnou experimentální přípravou. Jelikož byla příprava apoferritinových částic lenvatinibu neúspěšná,... Tyrosine kinase inhibitors (TKI) are small organic molecules designed for the targeted cancer therapy. They perform the inhibition of activated receptor tyrosine kinases in tumor cells, that defeats tumor growth, proliferation, metastasis and angiogenesis in tumor tissue. Two TKI, lenvatinib and vandetanib, are used in thyroid cancer treatment. This thesis investigates the ways leading to enhancement of efficiency of these anticancer drugs for therapy. One of the studied anticancer drug - lenvatinib - was investigated to be prepared in a nanoform. Nanoparticles were based on protein apoferritin as well as on lipids. Theoretical model of lenvatinib interaction with an apoferritin cavity, as well as the model of its encapsulation obtained by computer modeling indicated that lenvatinib seems not to be suitable for preparation of apoferritin nanoparticles. Since lenvatinib occurs in its neutral form during preparation of nanoparticles, it does not interact with nanoparticle. The unsuccessful experimental preparation of lenvatinib-loaded apoferritin nanoparticles confirmed that lenvatinib is not suitable for its preparation.
