Abstrakt: [cze][eng] NK buňky a KIR receptory: jejich význam v transplantacích hematopoetických kmenových buněk u leukemií Abstrakt Tato bakalářská práce pojednává o významu NK (natural killer) buněk u leukémií. Je zaměřena na strukturu a reaktivitu NK buněk, a především pak na transmembránové KIR receptory (killer cell immunoglobuline-like receptors) NK buněk, které hrají důležitou roli v likvidaci leukemických buněk (reakci štěpu proti leukemii, GvL) a tedy v celkové prognóze nemoci. Aktivační a inhibiční receptory KIR svou kooperací řídí cytotoxickou aktivitu NK buněk. Typizací genů pro KIR receptory u dárců hematopoetických kmenových buněk tak lze předpovědět úspěšnost léčby. Vyšetření genů KIR se využívá především u pacientů diagnostikovaných akutní myeloidní leukémií. It focuses on the structure and reactivity of NK cells, and especially on transmembrane KIR receptors (killer cell immunoglobulin-like receptors) of NK cells, that play an important role in the elimination of leukemic cells (graft-versus leukemia reaction, GvL) and thus in the overall prognosis of the disease. Activation and inhibitory receptors of KIR, by their cooperation, control the cytotoxic activity of NK cells. Thus, the typing of KIR receptor genes in hematopoietic stem cell donors can predict treatment success. KIR genes examination is mainly used in patients diagnosed with acute myeloid leukemia.
