Přestože podíl řepky olejky (Brassica napus) vůči ostatním olejnatým plodinám na světovém trhu neustále stoupá, její potenciál pro kultivaci je, tak jako u řady dalších plodin, stále omezený kvůli její citlivosti na abiotický stres chladem. Teoretická část této práce se proto zabývá problematikou rostlinného stresu a popisem metody RT-qPCR v kontextu vhodnosti jejího užití pro kvantifikaci změn ke kterým během chladového stresu dochází. V praktické části byla provedena analýza genové exprese u vybraného dehydrinového genu ERD10 včetně popisu odebírání vzorků z rostlin, způsob izolace RNA, reverzní transkripce do cDNA a vlastní RT-qPCR. Výzkumný soubor tvořili dvě odrůdy řepky Cadeli a Navajo podrobené stresu chladem při (2°)C. Původním předpokladem bylo, že mezi těmito odrůdami bude znatelný rozdíl v úrovni exprese ERD10 genu v závislosti na schopnosti těchto rostlin odolávat stresu chladem. Interpretace dat pak i přes jejich neúplnost tuto původní domněnku potvrdila. V průběhu prvních 6ti dní stresu chladem docházelo u odrůdy Navajo k znatelně vyšší expresi oproti odrůdě Cadeli. Na základě souvislosti mezi expresí ERD10 a odolností vůči chladu by mělo být možné použít ERD10 jako jeden z markerů k selekci chladu odolných genotypů. Although the share of rapeseed (Brassica napus) in comparison with other oil crops on the world market continues to rise, its cultivation potential ,as with many other crops, is still limited due to its abiotic cold stress sensitivity. Therefore, the theoretical part of this thesis deals with the issue of plant stress and the description of the RT-qPCR method in the context of its suitability for quantification of changes that occur during cold stress. In the practical part of this thesis, an analysis of gene expression in selected dehydrin gene ERD10 was performed including description of sampling from plants, method of RNA isolation, reverse transcription into cDNA and own RT-qPCR. The research sample consisted of two rapeseed varieties Cadeli and Navajo subjected to cold stress at (2°C). The initial assumption was that there would be a noticeable difference in the expression level of the gene ERD10 between these varieties, depending on the ability of these plants to withstand cold stress. Although some samples were not measured, the interpretation of the data confirmed original assumption. During the first 6 days of cold stress, the Navajo variety had a significantly higher expression of this gene than the Cadeli variety. Based on the association between ERD10 expression and cold resistance, it should be possible to use ERD10 as one of the markers for the selection of cold resistant genotypes.
