[cze][eng] Prionový protein (PrPC) je spojen především s původem transmisivních spongiformních encefalopatií (TSE), smrtelných onemocnění, jejichž molekulární podstatou je konverze buněčné formy proteinu PrPC na infekční formu PrPTSE. Ta je odolná vůči proteázám a běžným dekontaminačním metodám, hromadí se v tkáních a způsobuje degenerativní poškození centrální nervové soustavy. Potenciální fyziologická funkce PrPC v buňkách organismů zůstává nejasná, i když výzkumu tohoto tématu bylo v posledních letech věnováno značné úsilí. Exprese PrPC byla detekována především v nervových buňkách, vysoké hladiny proteinu se však vyskytují i v různých buňkách imunitního systému. Zatímco některé imunokompetentní buňky byly již široce zkoumány, jiné zůstávají z hlediska funkce PrPC nepoznány. PrPC zřejmě hraje roli při diferenciaci a aktivaci řady typů imunitních buněk, podílí se na regulaci produkce cytokinů a dalších imunitních procesů, ovlivňuje růst populace CD4+ T-buněk a také utváření sekundárních lymfatických orgánů. V této bakalářské práci byly shrnuty dosavadní poznatky z oblasti výzkumu úlohy PrPC v imunitních buňkách, jež je důležitý i z hlediska výzkumu diagnostiky a léčby prionových chorob. Klíčová slova: prionový protein, PrPC, PrPTSE, prionové choroby, imunitní systém, imunokompetentní buňky, fagocyty,...Prion protein (PrPC) is connected with the origin of transmissive spongiform encephalopathies (TSEs), fatal diseases that are on the molecular level based on the conversion of the cellular form of prion protein, PrPC, into the infectious form, PrPTSE. This isoform, exhibiting increased resistance against proteases and common decontamination methods, accumulates in tissues and causes degenerative damages of the central nervous system. Potential physiological function of PrPC in cells remains unclear, though many efforts have been focused on this research area in past years. Expression of PrPC was detected especially in neurons, high levels of PrPC are also present in different types of cells of immune system. Whereas some immunocompetent cells were widely examined, the relationship of PrPC with the function of others was not studied. PrPC probably plays a role in differentiation and activation of some immune cells, participates in regulation of cytokine production and other immune processes, affects grow of CD4+ T-cell population and also takes a part in formation of secondary lymphatic organs.
