[cze][eng] Elektrické signály jsou charakteristickým znakem nervové aktivity. K tomu, abychom pochopili fungování mozku, musíme být schopni sledovat tyto elektrické signály. Slibný způsob sledování elektrických signálů v neuronech zahrnuje použití fluorescentního proteinu citlivého na napětí a optický mikroskop. Jeden z nejlepších fluorescentních proteinů citlivých na napětí je ArcLight. Cílem této práce bylo zjistit molekulární mechanismus funkce tohoto konstruktu. Nejdříve jsou v této práci popisovány metody, které se zabývají měřením membránového potenciálu a s ním spojených změn intenzity fluorescence. V poslední části jsou uvedeny výsledky našeho mikroskopického pozorování.Electrical signals are a hallmark of neuronal aktivity. In order to understand how the brain works, we need to be able to visualize the electrical signals. A promising way to observe the electrical signals in neurons involves using a voltage sensitive fluorescent protein and an optical microscope. One of the best voltage sensitive fluorescent proteins is ArcLight. The aim of this work was to investigate the molecular mechanism of the function of this construct. First, this work describes methods for measuring membrane potential and associated changes in fluorescence intensity. The last part presents the results of our microscopic observation.
