Exprese rekombinantních proteinů v semenech je výhodná zejména kvůli vysoké koncentraci zásobních proteinů v malém objemu. Rekombinantní proteiny se v semenech akumulují ve vysokých koncentracích, zůstávají stabilní řadu let a díky nízkému obsahu alkaloidů a dalších látek nejsou semena náročná na další zpracování. Mezi další výhody řadíme minimální riziko kontaminace. Bakalářská práce se zabývá testováním transientní exprese genových konstruktů přímo v dozrávajících semenech sóji. V rámci této práce byly připraveny genové konstrukty pro testování transientní exprese genu uidA. Také byl studován vliv aditiv acetosyringonu a dithiothreitolu a jejich kombinací na transientní expresi a aktivitu genu uidA. Dále byla studována optimální délka a prostředí sonikace. Na závěr byla také porovnána účinnost transientní exprese metodou in vitro a in vivo. Bylo potvrzeno, že acetosyringon má pozitivní vliv pouze na semena, která nebyla předem sonikována. S prodlužující se sonikací klesá relativní aktivita genu uidA. Dále byl zjištěn pozitivní vliv dithiothreitolu u nesonikovaných semen. Vyšší hodnoty sledované relativní aktivity genu uidA byly také zaznamenány po přidání obou aditiv do infiltračního pufru. Jako optimální délka sonikace byla zjištěna 30 s sonikace v případě metody in vitro a 60 s u in vivo... The expression of recombinant proteins in seeds is especially advantageous due to the high concentration of storage proteins in a small volume. Recombinant proteins accumulate in seeds at high levels, remain stable for many years, and because of the low content of alkaloids and other substances in seed environment they can be easily extracted. There is also a minimal risk of contamination. This bachelor thesis deals with the testing of transient expression of gene constructs directly in maturing soybean seeds. In this work, we have have prepared gene constructs for strong transient expression of the uidA gene in plant tissues. The impact of the acetosyringone and dithiothreitol additives and the impact of their combinations on the transient expression and activity of the uidA gene have been studied as well. Furthermore, the optimal length and environment of sonication have been studied. Finally, the effectiveness of transient expression by in vitro and in vivo has also been compared. It has been confirmed that acetosyringone has a positive effect only on seeds that had not been previously sonicated. With prolonged sonication, the relative activity of the uidA gene decreases.
