Název: Stanovení syntetických antioxidantů pomocí HPLC s elektrochemickou detekcí na detektoru z uhlíkové plsti Autor: Martin Baroch Vedoucí bakalářské práce: RNDr. Hana Dejmková, Ph.D. Katedra: Katedra analytické chemie V této práci bylo cílem nalézt vhodné podmínky pro HPLC separaci s následnou coulometrickou detekci na detektoru z uhlíkové plsti. Stanovovanými látkami byly syntetické fenolické antioxidanty, konkrétně propylgallát (PG), terciální butylhydrochinon (tBHQ), butylhydroxyanisol (BHA) a butylhydroxytoluen (BHT). Pro úplné oddělení analytů bylo nutné použít gradientovou eluci. Jako mobilní fáze byl použit methanol a acetátovo-fosfátový pufr o pH 4,5. Obsah methanolu v mobilní fázi se během 8 minut lineárně zvyšoval z 55 % na 95 % a následně byl tento obsah držen 2 minuty. Jako stacionární fázi byla použita kolona Lichrospher® RP-18 (125×4 mm, 5 µm) (LichroCART). Potenciály vkládané na pracovní elektrodu z uhlíkové plsti byly 1,40 V pro BHT a 0,80 V pro ostatní látky. Dva potenciály byly zvoleny pro lepší odezvu detektoru na BHT. Závislost velikosti signálu na koncentraci byla sledována v koncentračním rozmezí 1·10-4 - 1·10-6 mol dm-3 pro každý analyt. V tomto rozmezí jsou kalibrační závislosti lineární. Nepřesná měření byla získána u BHT. To může být, mimo jiné, zapříčiněno právě... The analytes were synthetic phenolic antioxidants, specifically propyl gallate (PG), tert-Butylhydroquinone (tBHQ), butylated hydroxyanisole (BHA) and butylhydroxytoluene (BHT). For the complete separation of the analytes, gradient elution was required. As a mobile phase methanol and acetate-phosphate buffer of pH 4.5 were used. Concentration of methanol in mobile phase was linearly increased from 55 % to 95 % during 8 minutes and next 2 minutes was concentration of methanol held at 95 %. As a stationary phase was used Lichrospher® RP- 18 (125×4 mm, 5 µm) (LichroCART) column. Potentials putted on a carbon-fiber working electrode were 1.40 V for BHT and 0.80 V for other analytes. Two potentials were chosen for a better detector response to BHT. The calibration dependence of the signal on the concentration was observed in the concentration range of · 10-4 - 1 · 10-6 mol dm-3 for all analytes. In this range the dependencies are linear.
